In this dissertation, the HPLC-ESI-MS and MS/MS technology were used to separate and identify saponins in panax notoginseng leaves with a proper analytical condition. Besides, notoginsenoside Fa was isolated from notoginseng folium with a determined optimal technology by using HPLC.HPLC-MS technology was a very important method to identify the components of Chinese traditional medicines. A reversed-phase high performance liquid chromatographic method with gradient elution (aqueous 1mM NH4OAc, buffered to pH=7 with NH4OH-acetonitrile) was performed in this work and 12 kinds of saponins were separated. the MS and MS/MS technology of the separated saponins were investigated and the saponins were identified as ginsenoside Rg1, Rb1, Rb2, Rb3, Rd, Rc and it's isomer, notoginsenosides R1, Fa, Fc, Fe and gypenoside IX.Regarding notoginseng folium as materials and notoginsenoside Fa as the product, reverse-phase high performance liquid chromatography was used to purify notoginsenoside Fa. The effects of quantity of raw materials, the ratio and the flow rate of the mobile phase in the separation process were investigated respectively, and thus the suitable chromatography condition was determined: the mobile phase was composed of methanol and water with a ratio of 70:30, the flow rate of mobile phase was 30ml/min and the quantity of raw materials was 0.3~0.4g. Under the conditions established above, the content of notoginsenoside Fa was above 91%.
