Active components in traditional Chinese medicine (TCM) are its substance foundations for use in curing diseases. The studies of these active components in the drug therapies, toxicology and pharmacokinetics are important for clarifying the treatment mechanism of TCM, evaluating its security and controlling product quality.However, the chemical study of the active components in Chinese herbs and natural products is the basis of pharmacology research. In the study of component chemistry, the separation, purification, and structure identification of the active constituents, and the technology of mass production, and also the simultaneous separation and determination of these active constituents are the key points to the clarification of the active constituents and effective sites. And those are being hotspots.From the reference to the former work of our laboratory, in this paper, we have carried out some researches of new methodology in separation and purification of the active constituents, and in the simultaneous separation and determination of the active constituents in compound preparation on the basis of the summarization of the new developments at these aspects. And some significant results have been gotten. The main contents and results of this paper include:1. In the first section, we have made some researches on thetechnologic parameters of separation and purification of different types of ginsenosides with macroporous adsorption resin. The separation and purification of 20(s)-protopanaxtriol and 20(s)-protopanaxdiol type ginsenosides have been accomplished by eluting with the solution mostly containing water. The traditional separation and purification technology based on silica gel column need consume much time and toxic organic solvent. However, this new method has the merits of low consume and low pollution. So it can provide more suitable raw material processing method not only for ginsenosides Rg3 and Rh2, which are. high therapeutic effect for cancer, but also for the further study of the effective sites of ginseng.2. In the traditional techniques of separation and purification of active constituents in TCM, one of the main disadvantages is the large solvent and time consume, which would increase the production cost. In this paper, we first introduced microwave into the area of separation and purification. A novel method combining macroporous resin adsorption with microwave assisted desorption (MAD) for separation of 20(S)-protopanaxtriol type ginsenosides and 20(S)-protopanaxdiol type ginsenosides has been developed. After saponins from ginseng root extract has been adsorbed on macroporous adsorption resin, the separation of 20(S)-protopanaxtriol type and 20(S)-protopanaxdiol type ginsenosides from the resin can be completed under different solvents and...
