Purpose: This study will analysis of the existing genetic data of the The National Center for Biotechnology Information(NCBI)'s GEO database and The Cancer Genome Atlas(TCGA)database in order to find key genes for cervical lymph node metastasis of papillary thyroid carcinoma(PTC)occurrence and new therapeutic targets.Methods: Data search was performed on the GEO database.A series of PTC tissues,paracancerous tissues,and neck metastatic lymph node tissue of PTC were collected and integrated.And R software was used to make differentially expressed gene analysis,to screen differentially expressed genes between PTC and paracarcinoma tissues(differentially expressed gene set A)and differentially expressed genes between PTC and cervical metastatic lymph nodes(differentially expressed gene set B).The differentially expressed gene set C(hub gene)of the intersection of differentially expressed gene set A and differentially expressed gene set B was further screened.The gene expression data of the hub gene in PTC tissues,paracancerous tissues,and cervical lymph node metastatic cancer tissues were extracted from the GEO database,and box diagrams were displayed,and the protein-protein interaction network analysis was performed on the proteins encoded by the differentially expressed genes.The expression data of hub genes related to cervical metastatic lymph nodes of PTC neck were extracted from the TCGA database,and the gene expression differences between cancer tissues and paracancerous tissues were compared.Univariate and multivariate logistic regression analysis were used to screen genes related to cervical lymph node metastasis of PTC.Then,survival analysis of independent risk genes was performed in the c Bio Portal database.Results: By searching the GEO database,the gene chip GSE60542 with PTC was obtained.GSE60542 contains 33 PTC samples,30 paracancerous samples,and 23 cervical lymph node metastasis of PTC samples.By analyzing the GSE60542 chip data,there were 704 differentially expressed genes in differentially expressed gene set A and 71 differentially expressed genes in differentially expressed gene set B.There are 9 hub genes(PRSS2,LIPG PLEKHH1,SORD,CITED1,CCL21,SLC26A4,EGFEM1 P,UBE2QL1)in the differentially expressed genes set C.The box plot results showed that the expression levels of the genes LIPG,PLEKHH1,SORD,and SLC26A4 were gradually reduced in the paracancerous tissues,PTC tissues,and cervical lymph node metastatic cancer tissues.The expression levels of 9 hub genes in PTC were extracted from the TCGA database.The results showed that the genes LIPG,PLEKHH1,SORD,and SLC26A4 showed low expression in PTC tissues.Multivariate regression analysis results showed: SLC26A4 is an independent risk gene for cervical lymph node metastasis in PTC.Through the c Bio Portal online analysis website,the gene SLC26A4,which has a greater effect on the survival time of patients,was initially screened.Analysis of protein-protein interaction network for proteins encoded by differentially expressed genes showed that there was an interaction network between SLC26A4 and the proteins encoded by TG,TSHR,and FCRL3 genes.Conclusion: The GEO and TCGA databases were used for chip data retrieval and systematic analysis of differentially expressed genes between PTC tissues,paracancerous tissues,and cervical lymph node metastasis of PTC.It was found that the SLC26A4 gene is closely related to the occurrence of PTC cervical lymph node metastases.The mechanism of metastasis provides new potential loci.
