Objective: Thyroid carcinoma is the most common endocrine malignancy.PTC is the most common pathological type of all thyroid cancers.The incidence of papillary thyroid carcinoma(PTC)has increased significantly worldwide in recent decades,accounting for more than 90% of new thyroid cancers.With appropriate treatment,10-20% of patients relapse.There is a need to develop better methods for predicting PTC recurrence.In cancer,the expression pattern of SLC transporters has changed dramatically.Soluble carrier family(SLC family)plays an important role in cancer.The relationship between SLC family genes and PTC recurrence remains unclear.In addition,there is new evidence that SLC12A4 is functionally associated with cancer development.However,the biological function of SLC12A4 in PTC has not been determined.The aim of this study was to construct a SLC family gene model for predicting PTC recurrence and to explore the potential biological function of SLC12A4 in PTC.Methods: RNA-sequencing data and relevant clinical data in The Cancer Genome Atlas for 502 samples of PTC were analyzed to determine the prognostic value of soluble carrier family genes in PTC.We analyzed soluble carrier family gene expression and function in the samples.Clustering identified two clusters in the data.Risk characteristics were identified using LASSO and Univariate Cox regression analysis,which divided the patients into low and high-risk groups.In addition,pan-cancer analysis was performed on the prognostic model,and the potential biological function of SLC12A4 in PTC was analyzed by bio-informatics.Results:The expression levels of 88 soluble carrier genes were significantly different in tumor tissues and normal tissues.The two PTC clusters had different distributions of gene expression.The expression levels of SFXN1,SLC12A4,SLC35A1,SLC35E1 and SLCO1C1 were markedly different between the two groups.The high risk and low risk groups had significant different prognoses(P< 0.05).Significant differences were identified for disease free survival(DFS),sex and T stage between the two subgroups.The risk score was identified as an independent prognostic variable(P< 0.05).In patients with PTC,solute carrier gene expression showed differential associations with clinicopathological variables.SLC12A4 is low expressed in PTC tissues and is associated with immune invasion.DNA methylation may act as an upstream regulator of SLC12A4 expression and biological function.Conclusion: In patients with PTC,solute carrier gene expression showed differential associations with clinicopathological variables.The five genes(SFXN1,SLC12A4,SLC35A1,SLC35E1,SLCO1C1)could be used as prognostic factors for PTC,particularly to predict PTC recurrence.The model can also predict the DFS of a variety of tumors well.SLC12A4 is a good prognostic biomarker for PTC.
