Purpose:To obtain the original expression data of hepatocellular carcinoma through the cancer genome atlas(TCGA),to screen the differentially expressed genes of hepatocellular carcinoma through the R language,to explore the pathogenesis of hepatocellular carcinoma,and to investigate the expression of POLQ protein in hepatocellular carcinoma.Through the clinical validation of immunohistochemistry,to analyze the relationship between POLQ protein and clinicopathological parameters of hepatocellular carcinoma and its clinical significance with the prognosis of hepatocellular carcinoma.Methods:1.The R language RTCGA toolkit was used to download the samples of hepatocellular carcinoma mRNA sequencing from the TCGA database to screening candidate genes.2.Immunohistochemistry was used to detect the expression of protein.Mann-Whitney U test and Spearman rank correlation were used to analyze the relationship between the expression of POLQ protein and clinicopathological parameters of patients with hepatocellular carcinoma;Kaplan-Meier was used to plot survival curves,and the effect of POLQ protein expression on the prognosis of patients with hepatocellular carcinoma was analyzed.Results:1.The original expression data of hepatocellular carcinoma from the TCGA database was comprehensively analyzed,and the target gene POLQ gene was obtained.The significance of the difference in the different expression levels of POLQ gene in different clinical data was analyzed.At the 0.05 level of significance,there were significant differences in the expression of POLQ gene in hepatocellular carcinoma tissues in different stages.2.In hepatocellular carcinoma tissues,the positive rate of the high expression of POLQ protein 41.2%was higher than that of paracancerous tissues 0%.which was statistically significant(x2=41.57,P<0.001);theexpression of POLQ protein was related to the number of hepatocellular carcinoma tumors,the risk of recurrence,ALT,GGT,and BCLC staging(P<0.05),and there were significant differences.There was no correlation with age,gender,cirrhotic nodules,pathological grade,Stage,cirrhosis,HBsAg,HBcAb,TB,ALB,AFP,etc(P>0.05).Conclusion:1.TCGA database and bioinformatics verify that the expression of POLQ gene in hepatocellular carcinoma tissues is significantly higher than that in paracancerous tissues;the high expression of POLQ gene indicates that patients with hepatocellular carcinoma have a late stage and a poor prognosis.2.Immunohistochemistry clinically verifies that the high expression level of POLQ protein in hepatocellular carcinoma tissues is significantly higher than that of paracancerous tissues.The expression of POLQ protein is positively correlated with the ALT,GGT,tumor number,recurrence risk,and staging of patients with hepatocellular carcinoma;the high expression of POLQ protein is negatively correlated with the survival time of patients with hepatocellular carcinoma.
