Objective To analyzethe change of microRNAs expression profile in the tissues of laryngeal squamouscell carcinoma(LSCC)and its metastastic cervical lymph nodes,and to explore the potential regulatory mechanism and clinical significance of microRNAs in metastastic process of LSCC.Methods 1.To collect the specimens of LSCC tissues,normal tissuec adjacent LSCC and cervical lymph nodes tissue from patients with LSCC,2 cases with metastastic cervical lymph nodes and 1 case without metastastic cervical lymph nodes.2.To observe the microRNA profile of LSCC tissues,normal tissues adjacent LSCC and cervical lymph nodes tissues using microRNA microarray technique(Agilent Human miRNA microarry),and to screen the differential expression microRNA in LSCC tissues(control group is normal tissues adiacent LSCC),LSCC tissues with meatastastic cervical lymph nodes(contral group is LSCC tissues without metastastic cervical lymph nodes)and metastastic cervical lymph nodes(control group is non-metastastic cervical lymph nodes)using bioinformatics analysis technique(Fold change value>2;P<0.05).3.To analyze screened differential expression microRNA by GO and KEGG analysis technique using DAVID softwareResults 1.Analyzing by mircoRNA microarray technique,the results showed that(1)in the tissues of LSCC(contrast with normal tissue adjacent LSCC),24 differential expression microRNAs were found(21 microRNAs upregulated and 3 downreguated,respectively),(2)in the tissues of LSCC with metastastic cervical lymph nodes(contrast with LSCC without metastastic cervical lymph nodes),63 differential expression microRNAs were found(33 microRNAs upregulated and 30microRNAs downregulated,respectively),and(3)in the tissues of metastastic cervical lymoh nodes(contrast with non-metastastic cervical lymph nodes),94 differential expression microRNAs were found(56 microRNAs upregulated and 38 microRNAs downregulated,respectively).2.Basen on the results analyzed by GO and KEGG function enrichcment analysis technique,the interaction networks were constructed among differential expression microRNAs,related candidate target genes and items of GO and KEGG.The results showed that(1)in the tissues of LSCC,significantly differentially expressed miR-9a-3p and miR-7-5p were found;(2)in the tissues of LSCC with metastastic cervical lymph nodes,significantle expressed miR-6847-5p and miR-182-5p were found;(3)in the tissues of metastastic cervical lymph nodes,significantly differentially expressed miR-6874-5p and miR-214-3p were found.These microRNAs involved in several biological processes(such as transcriptional regulation,regulating target genes,ect.)and some cell signal pathways(such as Wnt,mTOR,ect.).Conclusion 1.microRNAs may play an important role in the initiation of LSCC,and may regulate its occcurance at the genepost-transcriptional level.2.microRNAs may play an important role in the process of cervical lymph node metastasisin LSCC.Specific microRNAs may determine the metastatic ability of LSCCcells,andmay also indicate the existence of tumor cell subgroups with different metastatic capabilities in LSCC.
