BackgroundBladder cancer is one of the most common malignant tumors around the word.Patients suffer from bladder cancer had poor quality of life and overall survival time.The prognosis of patients with advanced bladder cancer is poor.With unclear etiology,limited treatment options can be applied.Therefore,reliable molecular markers and therapeutic targets for early detection and novel therapy targets for bladder cancer has long been a hot spot in bladder cancer research.In this research,we tried to determine the key genes that play important roles in the developing of bladder cancer according to GEO and TCGA database by applying R software and its related expansion packages.Exploring the molecular mechanism of related genes to provide a bioinformatics basis for searching the etiology and pathogeny of bladder cancer.MethodsFirst,GSE130598 was screened and downloaded from the PUBMED GEO database.Next,significantly differentially expressed genes between bladder cancer tissues and para-cancerous tissues were found out by using R software and its related expansion packages.The differential genes were then subjected to GO enrichment and KEGG pathway enrichment analysis using R software.Finally,the key differential genes were found out by R software after constructing the protein-protein interaction(PPI)network by using Significantly differentially expressed genes.Downloading bladder cancer gene expression data from TCGA database,Sorting and analyzing the data to find out significantly differentially expressed genes using R software.Obtaining shared differentially expressed genes by intersecting the differentially expressed genes in the TCGA database with the core differentially expressed genes in the GEO database.Identifying the target core genes to study by reviewing the literature.the target core genes were analyzed using the GEO database of differentially expressed genes.Identifying core differential genes which are connected with the target core genes by Cytoscape.Results1.One hundred and fifty two differentially expressed genes between tumor and normal groups were found by analysing GEO database gene chip GSE130598 and using R software and bioinformatics methods.And,We found that 79 genes were elevated and 73 genes were decreased respectively..2.The differential genes in GSE130598 data were mainly enriched in MAPK pathway,Ras pathway and complexine phosphorylation,serine/threonine phosphorylation etc by GO and KEGG enrichment analysis of differential genes.3.The EPHA3 gene was found to be associated with the development of bladder cancer by combining GEO and TCGA databases and using R software?bioinformatics methods.Conclusions1.The occurrence of bladder cancer is of RAS and MAPK signal pathway.2.The low expression of EPHA3 in bladder cancer may be a tumor suppressor gene,and the role of EPHA3 gene in the process of bladder cancer needs further study.
