Background:Head and neck squamous cell carcinoma(HNSCC)is squamous cell carcinoma occurring in multiple anatomical sites of the head and neck.There are approximately 500,000 new cases worldwide each year,among which the prognosis of advanced-stage is poor and the five-year survival rate is only about 50%.Laryngeal carcinoma ranks third among HNSCC.Smoking,alcohol abuse,and HPV infection are all risk factors for HNSCC.Current treatments mainly include surgical treatment and non-surgical treatment,such as radiotherapy and chemotherapy,molecularly targeted therapy,and immunotherapy.Studies on the gene,RNA,protein and molecular signaling pathways of HNSCC are increasing,but the biological role of autophagyrelated long non-coding RNA has not been clearly studied now.The role of autophagy in tumor is inherently related to the status of related genes.Genes in different states can regulate the process of autophagy to promote or inhibit the proliferation and invasion of tumors,while lncRNA can regulate the status of autophagy genes through the corresponding signal axis.Objective:The aim of this study was to identify the key autophagy-related lncRNA and pathways associated with HNSCC as well as laryngeal carcinoma and to further analyze its molecular mechanism and prognostic significance.Materials and Methods:In this study,the RNA-seq tertiary dataset of HNSCC from The Cancer Genome Atlas(TCGA)included biological information of 45 normal head and neck tissues and 501 HNSCC sample tissues,while there are 528 files of corresponding clinical information.As for laryngeal carcinoma,we download RNAseq tertiary dataset of 11 normal larynx tissues and 112 laryngeal carcinoma sample tissues and 117 files of corresponding clinical information from TCGAHNSC.Differentially autophagy-related lncRNA expressed were screened by R software,and though the univariate Cox regression analysis and multivariate Cox regression analysis,we reveal the role of autophagy-related lncRNA in HNSCC and laryngeal carcinoma and display in graph and table.The pathway enrichment analysis of autophagy-Related lncRNA was performed by Cytoscape software programs.Meanwhile,survival analysis,independent prognostic analysis,receiver operating characteristic curve,and clinical correlation analysis were conducted to demonstrate the influence of autophagy-related lncRNA on patient prognosis and the reliability of the survival model.Results:1.We used bioinformatics technology to screen out 12 autophagy-related lncRNA in HNSCC,among which,the lncRNA with tumor suppressor effect included AC040977.1,AL450992.2,AL357033.4,RAB11B-AS1,AL139158.2,AC108010.1,AC104083.1,while the lncRNA with tumor promoting effect included AP002884.1,LINC00958,AC010894.2,AC022031.2,respectively.AC103702.2,and other 12 autophagy-Related lncRNA in laryngeal carcinoma,among which,the lncRNA with tumor suppressor effects included AC009113.1,AC008280.3,AF001548.2,LINC00324,AL391244.1,while the lncRNA with tumor promoting effects included MYOSLID,AL121899.1,AC007405.3,UCA1,AC012640.2,DTX2P1-UPK3BP1PMS2P11,AL513318.2.2.In the results of prognosis analysis,risk score was an important factor affecting the survival prognosis of HNSCC or laryngeal carcinoma and the higher the risk score,the worse the prognosis.Age,tumor stage and T,N,M stage also influence prognosis to some extent.3.We performed Gene Ontology(GO)enrichment analysis,the Kyoto Encyclopedia Genomes of Genes and(KEGG)signaling pathway analysis,but no statistically significant results were obtained.Conclusion:Five most important autophagy-related lncRNA,including LINC00958?UCA1?MYOSLID,RAB11B-AS1?LINC00324,that could provide more reliable biomarkers for early diagnosis and individualized control of HNSCC were identified.
