Objective:This study aimed to analysis the expression level of Wnt-1 induced secreted protein 2(WISP-2)gene in the tumor tissues of patients with esophageal squamous cell carcinoma(ESCC)and its correlation with the clinicopathological parameters and prognosis in ESCC patients.We also explored the role and clinical significance of over-expressed WISP-2 in the development and progression of ESCC,which may provide more targets and theories for the personalized diagnosis and treatment of ESCC patients.Methods:The clinical data of 102 ESCC patients who received surgical treatment in our hospital from April 2012 to March 2016 were retrospectively analyzed.The expression levels of WISP-2 protein in tumor tissues(experimental group)and the normal adjacent tissues(control group)in paraffin specimens were detected by immunohistochemical staining.20 representative ESCC patients were further picked up from the parent and western blotting was used to detect the expression levels of WISP-2 protein in tumor tissues and the normal adjacent tissues.Meanwhile,the prognostic factors of ESCC patients were analyzed and the risk of Cox regression was evaluated.Results:1.The expression rate of WISP-2 protein in the carcinoma tissues of 102 ESCC patients was63.73%,significantly higher than that of 36.27% in the corresponding adjacent tissues(P< 0.05).2.The results of Western blotting showed that the average expression level of WISP-2protein in the cancer tissues of ESCC patients was significantly higher than that in the corresponding adjacent tissues(P<0.001),which further confirmed the high expression of WISP-2 protein in ESCC cancer tissues.3.There was no significant correlation between the expression level of WISP-2 protein and clinical parameters such as gender,age and tumor volume,but there was significant correlation between the expression level of WISP-2 protein and parameters such as tumor tissue invasion depth,clinical stages,differentiation degree and lymph node metastasis.4.The expression level of WISP-2 protein could be an independent risk factor affecting the overall survival rate of ESCC patients;patients with ESCC with low expression of WISP-2protein in cancer tissues had longer disease-free survival and overall survival than those with high expression of WISP-2(P<0.001).Conclusions:1.WISP-2 protein is highly expressed in ESCC carcinoma tissues,and the expression level of WISP-2 protein in carcinoma tissues is significantly higher than that in corresponding adjacent tissues,suggesting that WISP-2 gene may be involved in the occurrence and development of ESCC as pro-oncogenic factor.2.The highly expressed WISP-2 protein in cancer tissues is significantly correlated with the clinical stage,tumor tissue invasion depth,differentiation degree and lymph node metastasis of ESCC patients,which expected to be an important new therapeutic target and potential biomarker for prognosis assessment of ESCC patients.
