ObjectiveIn the study of modern meridians,the relationship between meridians and tissue fluid,tissue gap and fascial connective tissue is one of the important research ideas to link meridians with known tissue structure.The purpose of this study is to observe the distribution and morphology of the tissue fluid channel by means of fluorescence photography,and to observe the effect of blocking the tissue fluid channel of the meridian circulation site on the corresponding physiological function of rats,and to explore the correlation between blocking the low hydraulic resistance channel along meridian(LHRCM)and "channel blockage",so as to provide a new method and experimental basis for the study of meridian.Method1.Experiment 1:Observation of the low hydraulic resistance channel(LHRC)of rat's Conception Vessel(CV)with the method of fluorescence photography24 healthy adult male SD rats were randomly divided into three groups:low impedance point(LIP)group(n=8),low hydraulic resistance point(LHRP)group(n=8)and non-meridian points(NMP)group(n=8).After anesthesia,depilate the abdomen of rats,locate the low impedance line on the middle abdominal line of rats with meridian locator,and peel off the abdominal skin.In the LIP group,fluorescein sodium was injected into the LIP of the CV.In the LHRP group,fluorescein sodium was injected into the LHRP of the CV.In the NMP group,fluorescein sodium was injected into the high resistance point of the NMP.The migration and distribution of fluorescein sodium in each group were observed by fluorescence photography2.Experiment 2:the effect of ligation on the migration of sodium fluorescein21 healthy adult male SD rats aged 5-8 weeks,8 in conscious group,8 in ligation group and 5 in anesthesia group.One week before the experiment.the rats in the ligation group were used to ligate the low flow resistance channels along CV.Rats in anesthesia group were anesthetized with pentobarbital sodium and rats in conscious group and ligation group were anesthetized with mixed oxygen.The low impedance line along CV was measured by meridian locator,the LIP of CV was located by rheometer,and fluorescein sodium was injected at the LIP of CV.Rats in conscious group and anesthesia group were put back into feeding cage after injection of fluorescein sodium,and then they were anesthetized again 15 minutes later.The fluorescence migration images were collected by fluorescence photography 20 minutes after the injection of fluorescein sodium.Then compare and analyze the characteristics of the migration tracks of fluorescein sodium in the three groups3.Experiment 3:Observation on the effect of ligation of LHRC of CV on reproductive system24 4-week-old female SD rats were randomly divided into three groups:CV ligation group(CVL group),non-meridian ligation group(NML group)and control group,with 8 rats in each group.After anesthesia,the rats were depilated and the low resistance line along CV was measured.Rats in CVL group and NML group were ligated on the low impedance line of CV and the non-meridian part respectively.The body weight of rats was observed every week,the serum hormone level was measured in the fifth week after modeling,the organs were dissected and separated,and the organ coefficient was calculatedResult1.Experiment 1:(1)the characteristics of sodium fluorescein trace:in LIP group and LHRP group.the linear migration was observed along the direction of C V.In NMP group,no linear migration was observed.(2)The migration length of fluorescein sodium along the direction of CV:the average length of LIP and LHRP group was 1.88±0.31 cm and 1.73 ±0.26 cm,which were significantly longer than the average length 1.19±0.18cm of NMP group(P<0.01);the length of LHRP group was slightly shorter than that of LIP group,but the difference was not statistically significant(P>0.05).(3)The width of sodium fluorescein trace:the average width of LIP group was 0.71 ±0.15 cm,and that of LHRP group was 0.63±0.1 9 cm,which were significantly smaller than that of NMP group(1.21 ±0.28cm)(P<0.01);the width of LHRP group was slightly smaller than that of LIP group,but the difference was not statistically significant(P>0.05).(4)The ratio of length to width of sodium fluorescein trace:the average ratio of length to width of LIP group was 2.67 ±0.26,and that of LHRP group was 2.99±0.93,which were significantly higher than that of NMP group 1.02 ± 0.17(P<0.01).The ratio of length to width of LHRP group was slightly higher than that of LIP group,but the difference was not statistically significant(P>0.05).(5)The mean migration velocity of fluorescein sodium was 0.166±0.059 cm/min in LIP group and 0.135± 0.048 cm/min in LHRP group,which were significantly faster than that in NMP group 0.040± 0.037 cm/min(P<0.01);the migration velocity of fluorescein sodium of LHRP group was slightly smaller than that in LIP group,but there was no statistical difference(P>0.05)2.Experiment 2:(1)the migration length of sodium fluorescein along the direction of CV:the average length of conscious group was 4.81±0.45cm,significantly longer than that of anesthesia group 2.58±0.36 cm and ligation group 1.19±0.20 cm(P<0.01).(2)The width of sodium fluorescein track:the width of sodium fluorescein track in conscious group and ligation group was slightly smaller than that in anesthesia group,but there was no statistical difference(P>0.05).(3)The ratio of length to width of sodium fluorescein trace:the ratio of length to width of conscious group was 2.64±0.28,which was significantly higher than that of anesthesia group 1.10±0.10 and ligation group 1.15±0.10(P<0.01).3.Experiment 3:(1)body weight:the body weight of the rats in the CVL group was significantly lower than that in the NML group and the control group(P<0.01).(2)Serum E2,FSH,LH:compared with the control group,the serum E2,FSH,LH of the CVL group were significantly lower(P<0.05 or P<0.01),and the serum FSH of the NML group were significantly lower(P<0.01);compared with the NML group,the serum E2,FSH,LH of the CVL group were significantly lower(P<0.05 or P<0.01).(3)Organ weight coefficient compared with the control group,the weight coefficient of ovary and kidney in CVL group was significantly lower(P<0.05),the weight coefficient of spleen in CVL group was significantly higher(P<0.05),and the weight coefficient of spleen in NML group was significantly higher(P<0.05);compared with the NML group,the weight coefficient of spleen in CVL group was significantly higher(P<0.05)Conclusion1.Based on fluorescence photography,a new method of visualization observation of LHRCM was established,which provides a safe and convenient new method for the visualization of meridians.2.Using the method of visualization,the existence of the LHRC of rat's CV was observed.It was proved that the tissue fluid around CV flowed in the direction of the meridians rather than spread evenly,and the blocking effect of ligation on the LHRC of CV was verified.3.Ligation of the LHRC of CV can reduce the serum E2,LH and FSH levels.It suggests that blocking the LHRP of CV can affect the reproductive function of the rats.So that the LHRC of CV may be related to CV,which provides the experimental basis for the hypothesis of the low flow resistance channel of meridians.
