Objective: To investigate the effect of Zheng's No.3 fumigation on postoperative adhesions in TGF-?1 of adhesive tissue and IL-1 of the joint fluid after reconstruction of the anterior cruciate ligament of knee in New Zealand rabbits.It will further clarify the mechanism of external fumigation and washing of Zheng's No.3fumigation to treat postoperative adhesions of knee joint.Methods: 18 healthy New Zealand female rabbits were randomly divided into three groups: Zheng's No.3 fumigation group,saline fumigation control group and blank group.The reconstruction of anterior cruciate ligament(ACL)was performed by autologous Achilles tendon transplantation in the right knee of all rabbits after grouping.Plaster cast straight position fixed,left knee are not placed.After 3 days of operation,the knee joint was fixed with fixed plate at the same time,and the knee joint was fumigated after washing,and the plaster was fixed for 3 times per week for 4 weeks;blank group did not do fumigation but at the same time unfastened gypsum and fixed the plate fixed for the same time.the IL-1 content in knee joint fluid was detected by enzyme-linked immunosorbent assay(ELISA)after 4 weeks of execution of all rabbits,and the expression level in adhesion tissue TGF-?1 knee joint was detected by immunohistochemistry.By using statistical software SPSS 20.0 statistical description and inference,the mean and standard deviation are used for quantitative data expressed,the inter-group contrast was analyzed by one-factor analysis of variance,and the comparison among groups was compared by the least difference significant method(LSD method).the test level ?=0.05.when P<0.05,The difference was statistically significant.Results:1.After 4 weeks of treatment,the external fumigation of Zheng's No.3fumigation can effectively reduce the degree of adhesion and histopathological findings in the model after reconstruction of anterior cruciate ligament in New Zealand rabbits.2.The IL-1 level of knee joint fluid in rabbits was lower than that in normal saline group and blank group(P<0.01),but there was no significant difference between normal saline group and blank group(P>0.05);3.The TGF-?1 content of knee joint adhesion tissue decreased effectively(P < 0.01),there was no significant difference between the saline group and the blank group(P>0.05).Conclusion : 1.the external fumigation of Zheng's No.3 fumigation drug can effectively reduce the degree of joint cavity adhesion and improve the histopathological findings after knee ligament reconstruction in New Zealand rabbits.2.the external fumigation of Zheng's No.3 fumigation drug can reduce the IL-1 level in knee joint fluid of model rabbit operation,but the normal saline has no obvious effect.3.the external fumigation of Zheng's No.3 fumigation drug can effectively reduce the content of TGF-?1 in the adhesion tissue of the knee joint of the model rabbit,and the physiological saline has no obvious effect.4.The prevention and treatment of the adhesion of Zheng's No.3 fumigation drug to the model rabbit jointadhesion after anterior cruciate ligament reconstruction may be by inhibiting the inflammatory reaction and the formation of adhesion tissue.
