Purpose:1.Confirm the success of the selected modeling method,and preliminary explore the relationship between FGF21 and blood glucose,blood lipid and insulin resistance index2.To observe the effects of Shugan Huazhi method on NAFLD model,including the effects on liver cell steatosis and liver function.3.Investigate the possible mechanism of the treatment of NAFLD by Shugan Huazhi method,including protecting liver cells,improving lipid deposition,and its effect on FGF21 protein and gene expression.Material and method:A total of 60 SPF-grade SD rats were divided into 4 groups,of which 20 were in the normal group and 10 in the model group,and 10 were in the western medicine group and the Chinese medicine group.The normal group was given normal feed,the other groups were given high-fat feed,and the four groups maintained normal drinking water,while ensuring that the breeding environment was quiet and comfortable,and natural lighting was provided.After 8 weeks of feeding,10 rats in each of the normal group and the model group were taken.The serum and liver tissue of the rats were taken,and the serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),and triglyceride(TG)were measured according to the kit instructions.),Cholesterol(TC),low-density lipoprotein(LDL-C),high-density lipoprotein(HDL-C),and fibroblast growth factor(FGF)21.Hepatic tissue was stained with HE and observed to determine whether the modeling was successful.The remaining 40 rats stopped feeding with high-fat diet and started treatment after 8 weeks of feeding.The normal group and the model group were fed with saline,and the Chinese medicine group was given Shugan Huazhi Capsule Suspension.Gastrointestinal suspension is administered to the stomach.After 4 weeks,rat serum and liver tissues were taken,and relevant biochemical indicators such as blood glucose and blood lipid were detected;HE staining of liver tissue was performed to observe fatty changes of liver cells;western blot was used to detect FGF21 protein expression;real-time fluorescence quantification(real-time PCR)was used to detect FGF21 gene expression.All values are expressed as mean ± standard deviation(x ± s).SPSS 23.0 software was used for T test or analysis of variance(ANOVA).When p <0.05 inches,the difference was considered statistically significant.Results:1.Serum TG,TC,LDL-C,HDL-C test results of rats in each group: Compared with the model group,the serum TG,TC,LDL-C content of the Chinese medicine group was significantly reduced,and the difference was statistically significant(P <0.01)HDL-C content was significantly increased,and the difference was statistically significant(P <0.01).Compared with the Western medicine group,the TG,TC,and LDL-C contents in the serum of the Chinese medicine group were slightly reduced,but the difference was not statistically significant(P> 0.05),HDL-C content increased slightly,but the difference was not statistically significant(P> 0.05)2.Serum ALT and AST test results in each group of rats: Compared with the model group,the levels of ALT and AST in the serum of the Chinese medicine group were significantly reduced,and the difference was statistically significant(P <0.05);compared with the western medicine group,the serum of the Chinese medicine group The ALT content decreased slightly,but the difference was not statistically significant(P> 0.05).The AST in the serum of the Chinese medicine group was significantly reduced,and the difference was statistically significant(P <0.05).3.Pathological changes of liver in each group of rats: Compared with the model group,the liver surface of the rats in the Chinese medicine group was significantly improved,the surface was smooth,the color was lighter than the normal group,but it was still red,and there was no greasy feeling.It showed that the hepatocytes were relatively normal and relatively uniform,the arrangement of hepatocyte cords was normal,and the fat droplets in the liver were significantly reduced and scattered as compared with the model group.4.Comparison of IR index of rats in each group: Compared with the model group,the IR index of the Chinese medicine group was significantly reduced,and the difference was statistically significant(P <0.05).Compared with the western medicine group,the IR index of the Chinese medicine group was slightly increased.But the difference is not statistically significant(P> 0.05)5.Results of liver FGF21 m RNA expression in each group of rats: Compared with the model group,the expression of FGF21 m RNA in the Chinese medicine group increased more significantly,and the difference was statistically significant(P <0.01);compared with the model group,the FGF21 m RNA in the western medicine group There was no significant difference in expression.6.Results of FGF21 protein expression in rat liver of each group: Compared with the model group,the expression of FGF21 protein in the western medicine group did not change significantly,and the difference was not statistically significant.Compared with the model group,the expression of FGF21 protein in the Chinese medicine group increased.More obvious,the difference is statistically significant(P < 0.01)Conclusion:The Shugan Huazhi method has a significant intervention effect on the NAFLD rat model induced by high-fat diet.This method has obvious effects of improving insulin resistance,regulating lipid metabolism,and protecting liver cells.Its possible mechanism is to improve the fatty degeneration of NAFLD rats by increasing the expression of FGF21 m RNA.
