Purpose: To observe the effects of Shugan Huazhi Tongluo decoction on Tumor necrosis factor-?(TNF-?),Interleukin-18(IL-18),Anine aminotransferase(ALT),Glutamate transaminase(AST),Glutamyl transpeptidase(GGT),Alkaline phosphatase(ALP),Total bilirubin(TBIL),and Direct bilirubin(DBIL)in the serum of rats with non-alcoholic fatty hepatitis(NASH).To explore the effectiveness of Shugan Huazhi Tongluo decoction in NASH treatment.Method: Eighty-four Wistar rats were cleaned and weighed after 1 week of adaptive feeding.84 rats with appropriate weight were randomly divided into 6 groups,fourteen in each group,half male and female.6 groups were then specifically grouped as blank control group,model control group,silibinamine group,high dose group of traditional Chinese medicine compound,medium dose group of traditional Chinese medicine compound,low dose group of traditional Chinese medicine compound.The blank control group was given conventional feed,and the other groups were fed with high-fat diet(feed composition: 86.8% basal feed + 3% cholesterol + 10% lard + 0.2% propylthiouracil)and continuous modeling was performed for 12 weeks.The control group and model control group were intragastrically administered with an equal volume of distilled water,and the cilibinamine group was intragastrically administered with silinbine aqueous solution(3.125 mg/ml).The rats in the high dose group of traditional Chinese medicine compound,the medium dose group of traditional Chinese medicine compound,and the low dose of traditional Chinese medicine compound,were intragastrically administered with Shugan Huazhi Tongluo decoction,respectively(concentrations: 3.12g/ml,1.56g/ml,0.78g/ml).After 12 weeks the model was successful.Six groups of animals were fasted for 16 hours and then anaesthetized with 20% carbitol solution(5 ml/kg).Abdominal aorta was used to take blood,and contents of AST,ALT,GGT,ALP,TBIL,DBIL in serum were measured by an automatic biochemical analyzer.The contents of TNF-? and IL-18 in serum were determined by enzyme-linked immunosorbent assay(ELISA).Result:1 Compared with the model group,Shugan Huazhi Tongluo high/middle-dose groups and celibenamine group ALT?AST had statistically significant differences((P<0.05,P<0.01,P<0.01),Shugan Huazhi Tongluo low dose group had significant statistical differences(p>0.05).The serum ALT and AST were significantly higher in the cilibinamine tablets group and the high-dose group of Shuganhuazhitongluo prescription(p<0.05);there was no significant difference in serum ALT and AST between the celibenamine tablet group and the middle dose group of Shuganhuazhitongluo prescription(p>0.05).There was no significant difference n serum ALT and AST and middle does group in Shuganhuazhitongluo prescription high group(p>0.05).2 The serum GGT and ALP activity of the Cilibinamine group and high-dose group of Shugan Huazhi Tongluo decoction were significantly lower than that of the model group(p<0.01,p<0.05),Shugan Huazhi Tongluo low dose group had significant statistical differences(p>0.05).The serum GGT,ALP activity of middle/low dose group of Shugan Huazhi Tongluo decoction,was no significant difference from the model group(p>0.05).There was no significant difference in serum GGT and ALP levels between the silibinamine tablet group and the high-dose group of Shuganhuazhitongluo decoction(p>0.05).3 The serum levels of total bilirubin(TBIL)and direct bilirubin(DBIL)in the NASH rats of the model group were significantly higher than those in the serum of the Shugan Huazhi Tongluo decoction high and middle dose group and the Cailinamine group(P<0.01?P<0.01?P<0.05),Shugan Huazhi Tongluo low dose group had significant statistical differences(p>0.05).The levels of TBIL and DBIL in the study were significantly lower than those in the control group(p<0.01).There was no statistical difference between the serum of cililbinamine tablets and TBIL and DBIL in the serum of high-and middle-dose groups of Shuganhuazhitongluo decoction(p>0.05),Compared with the middle-dose group,serum TBIL?DBIL in the high-dose group of Shuganhuazhitongluo prescription was significantly lower than that of the middle-dose group(p<0.01).4 The expression of TNF-? and IL-18 in the model group was significantly higher than that in the blank control group.Compared with the model group,TNF-? and IL-18 in the high and middle dose group and celibenamine group were significantly higher than those in the model group(P<0.01?P<0.01?P<0.05),there was no statistical difference between TNF-? and IL-18 in serum of Shuganhuazhitongluo low-dose group and model group(p>0.05);The serum levels of TNF-? and IL-18 in the silibinamine tablets group were significantly higher than those in the high-dose group of Shuganhuazhitongluo decoction,with statistically significant differences(P<0.01),there was no difference between the silibinamine tablet group and the middle dose group of Shug anhuazhitongluo decoction(P>0.05);there was no significant difference in serum TNF-? and IL-18 contents between Shu gan hua zhi tong luo decoction high-dose group and middle-dose group of Shu gan hua zhi Tong luo Decoction(P>0.05)there was no significant difference in serum TNF-? and IL-18 contents between high-dose group and middle-dose group of Shuganhuazhitongluo decoction(P>0.05).In conclusion:1 Shugan Huazhi Tongluo decoction can effectively improve liver biochemical indicators and reduce inflammation.2 Shugan Huazhi Tongluo decoction can effectively reduce the levels of TNF-? and IL-18 and relieve inflammation.
