| Purpose:Based on the theory of "liver intestine axis",the therapeutic effect of Shugan Huoxue Huazhi meth od on nonalcoholic fatty liver disease(NAFLD)model rats was observed,and the mechanism of Shugan Huoxue Huayi method on NAFLD model rats was analyzed from the changes of LBP and TBA related indexes of liver intestine circulation,so as to provide a therapeutic idea and strategies for clinical application of Shugan Huoxue Huayi method in the prevention and treatment of NAFLD.Material and method:Forty healthy male SD rats were fed with SPF for 7 days,then according to the weight of rats,the rats were divided into four groups: normal group,high fat model group,Chinese medicine treatment group,polyene phosphatidylcholine group,10 rats in each group.Among them,the normal group rats were fed with the basic diet of common diet,the other three groups rats were fed with high-fat diet.During this period,the four groups of rats were in the same feeding environment,and the temperature and humidity were suitable.After 56 days,the rats in each group were administrated with drugs,the volume of which was 1ml / 100 g.The normal group and the high fat model group were administrated with normal saline,the traditional Chinese medicine group was administrated with compound suspension of traditional Chinese medicine,the polyene phosphatidylcholine group was administrated with polyene phosphatidylcholine.The dosage of the traditional Chinese medicine group and the polyene phosphatidylcholine group was converted according to the conversion formula of dosage of human and rats,the formula was: one day's demand Crude drug amount / 70 kg × animal weight × 6.3.The times of gavage were once in the morning and once in the evening.The administration lasted for 28 days.At the end of 28 d,all rats in the four groups were killed,and serum and liver tissue were collected for the detection of various indexes.The contents of serum TC,TG,HDL-C,LDL-C,ALT and AST were detected by automatic biochemical analyzer;the contents of LBP and TBA were detected by enzyme-linked immunosorbent assay(ELISA);the pathological changes of liver tissue were observed by HE staining.Results:1.Compared with NCD group:The serum TG,TC,LDL-C,ALT and AST in HFD group were significantly increased,while HDL-C was significantly decreased(P <0.01);The morphology of hepatocytes was obviously swollen,the arrangement of cells was not orderly,there were many round fat drop vacuoles with different sizes,the location of nuclei was shifted,the inflammatory cells infiltrated in different degrees in the lobule and the portal area of the liver,and the overall characteristics of lipid degeneration were obvious;LBP and TBA of liver intestine circulation increased(P <0.05).2.Compared with HFD group:The levels of TG,TC,LDL-C,alt,AST and HDL-C in serum of rats in Shugan Huoxue Huayu group were significantly lower than those in the control group(P <0.05);The swelling state of hepatocytes was obviously improved,the lipid drop was significantly reduced,and the arrangement of cell cords was significantly reduced compared with the regular hepatocytes;LBP and TBA of liver intestine circulation decreased(P <0.01).3.Compared with western medicine group:The levels of TG,TC,LDL-C,alt,AST and HDL-C in serum of rats in Shugan Huoxue Huayu group were all decreased,but the levels of HDL-C were increased,but the difference was not statistically significant(P> 0.05);There was no significant difference in liver pathology;LBP and TBA decreased,but there was no statistical significance(P> 0.05).Conclusion:1.Shugan Huoxue Huazhi method can reduce the expression of LBP and TBA in the serum of NAFLD model rats,which indicates that Shugan Huoxue Huayi method can reduce the level of LBP and TBA by regulating the circulation of liver and intestine,and achieve the effect of treating NAFLD.2.Shugan Huoxue Huazhi method can reduce the levels of ALT,AST,TG,TC,LDL-C and HDL-C,which shows that Shugan Huoxue Huayi method can improve the liver injury and lipid metabolism. |
