Objective:By establishing a rat model of mycoplasma pneumonia,the effect of the mycoplasma mixture on the lung histopathology,IFN-?,and IL-4 factors of the mycoplasma pneumonia rat was observed,and the therapeutic effect and mechanism of the mycoplasma mixture on mycoplasma pneumonia were explored.Methods :Forty-eight SPF male Wistar rats,weighing 200 ±15g,were fed adaptively for 1 week,and were divided into 4 groups according to the random number table method: blank group,model group,azithromycin group,and Zhiyan mixture group,12 rats in each group.Inoculation method,except the normal group,the remaining groups were inoculated with 100 ul of MP bacterial solution daily at a concentration of 106 CCU/ml for 4 consecutive days to establish a Mycoplasma pneumoniae pneumonia model.The normal group was given the same amount of liquid culture without MP Base,for 4consecutive days.Administration method: The blank group and model group were given intragastric saline,2ml each time,once a day.Azithromycin granules are administered on the 5th day,once a day,on the first day at a dose of 50 mg/kg,and on the next 4 days at a dose of 25 mg/kg,mixed with physiological saline into 2 ml of liquid,followed by continuous intragastric administration for 5 days.With normal saline.Zhiyan mixture group was given intragastric administration of Zhiyan mixture,2ml each time,once a day.After continuous intragastric administration for 10 days,6 rats were randomly selected from each group for intraperitoneal anesthesia after 5 days of intragastric administration.The rats were sacrificed by cardiac blood sampling.Lung tissues were harvested for HE staining pathological examination.Serum was taken to determine theexpression of IFN-? and IL-4 by ELISA,repeat the above operation after 10 days of gavage in the remaining rats.Results :1.HE staining results showed that the lung tissue structure of the blank group was intact,the alveolar wall was smooth and intact,and there was no obvious inflammatory cell infiltration.The lung tissue of the model group showed a large number of inflammatory cell infiltration,and the alveolar wall was significantly thickened,broken,and bronchitis In the mixture group and azithromycin group,lung tissue structure was damaged,some alveolar walls thickened and broken,and a small amount of inflammatory cells infiltrated.The pathological changes of lung tissue on day 10 of the four groups of rats were not significantly different from those on day 5.2.Pulmonary pathological tissue score results showed that: on the 5th day of intervention,the lung tissue pathology scores of the blank group and the model group were increased and there was a significant difference(P<0.01);the lung tissues of the azithromycin group,the Zhiyan mixture group and the blank group were compared The pathological score was elevated and there was a significant difference(P<0.01).Compared with the model group,the azithromycin group and the Zhiyan mixture group had lower lung histopathology scores(P<0.01);the azithromycin group and the Zhiyan mixture group There was no significant difference in comparison(P>0.05).On the 10 th day of intervention,there was no significant difference between the four groups(P>0.05).3.ELISA results showed that: on the 5th day of intervention,compared with the blank group,the expression of IFN-? in the model group was significantly reduced(P<0.01),and on the 10 th day of intervention,compared with the mod el group,the azithromycin group,Zhiyan mixture group The expression of IFN-? was significantly increased.There was no significant difference between the Zhiyanmixture group and the azithromycin group during the entire observation period(P>0.05).Compared within the four groups,there was no significant difference between the 5th and 10 th days of intervention(P>0.05).On the 5th day of intervention,compared with the blank group,IL-4 expression was significantly increased in the model group(P<0.01).On the 10 th day of intervention,compared with the model group,the IL-4 expression of the azithromycin group and the Zhiyan mixture group was significantly reduced(P<0.01),compared between the Zhiyan mixture group and the azithromycin group,there was no significant difference during the entire observation period(P>0.05),compared within the four groups,there was no significant difference between the 5th and 10 th day of intervention(P>0.05).Conclusion :1.Zhiyan mixture can improve the general condition of MP-infected rats and reduce inflammation of lung tissue.2.The expression of IFN-? decreases in mycoplasma pneumonia rats,and the expression of IL-4 increases in mycoplasma pneumonia rats.3.Zhiyan mixture can increase IFN-? expression of mycoplasma pneumonia uncle and reduce IL-4 expression.The mechanism of treatment of mycoplasma pneumonia may be related to the regulation of TH1/TH2 balance.
