Objective:To investigate the effect of zhiyan mixture on the expression of TLR4 and TNF-?in rats infected with mycoplasma pneumonia.Methods:48 SPF Wistar rats,male and weighing(200±15)g,were randomly divided into 4 groups:blank group,model group,azithromycin group and Zhiyan mixture group,12 rats in each group.The model group,azithromycin group and Zhiyan mixture group were dripped with 1*10 ~6CCU/ml MP strain 100ul for four consecutive days to establish mycoplasma pneumonia model.The blank group received nasal drops with the same amount of normal saline.After the model was established,the blank group and the model group were given normal saline,2ml each time,once a day.Azithromycin granules were administered for 5 days,with a dose of 50mg/kg on the first day and a dose of 25mg/kg on the fourth day.All the azithromycin granules were mixed with physiological saline to form 2ml of liquid,which was infused into the stomach continuously for 5 days.After 5 days,the same dose of physiological saline was continued to be infused into the stomach.Zhiyan mixture group was given Zhiyan mixture orally,2ml each time,once a day.Six rats in each group were anesthetized with 10%chloral hydrate after continuous gastric perfusion for 10 days.Blood was collected from the heart.After standing and centrifuging,the concentration of TLR4 and TNF-?were detected by Elisa.Lung tissues were taken for HE staining to observe pathological changes and histopathological scores.The remaining rats were gavaged for 10 days and the above operations were repeated.Results:1.HE staining results show that:in the model group,a large number of inflammatory cell infiltration can be seen,alveolar walls are obviously thickened and fractured;in the blank group,lung tissue structure is intact,alveolar walls are smooth and complete,and no obvious inflammatory cell infiltration is found;in the Zhiyanmixture group and azithromycin group,lung tissue structure damage can also be seen,some alveolar walls are thickened and fractured,and a small amount of inflammatory cell infiltration is found.There was no significant difference in pathological changes between thefour groups of rats on the 10th day and the 5th day.2.pathological tissue score results showed that:on the 5th day of intervention,the pathological score of the blank group and themodel group were different(p<0.01).the pathological damage of the model group was more serious than that of the blank group,and the score of the blank group was lower and the lung tissue was moreintact.The pathological score of azithromycin and Zhiyan mixture group was different from that of blank group(p<0.01),and alsodifferent from that of model group(p<0.01).There was nosignificant difference between azithromycin and Zhiyan mixture(p>0.05).On the 10th day of intervention,there was no significant difference among the four groups.3.Elisa results showed that on the 5th day after intervention,TLR4 and TNF-?in model group were higher than those in blank group(p<0.01).azithromycin group and Zhiyan mixture group were higher than those in blank group(p<0.01),lower than those in model group(p<0.01).there was no significant difference betweenazithromycin group and Zhiyan mixture group(p>0.05).On the 10th day after the intervention,there was no significant differencebetween the two-stage model groups(p>0.05),there was nosignificant difference between azithromycin groups(p>0.05),there was difference between Zhiyan mixture groups(p<0.01),and there was difference between Zhiyan mixture and azithromycin groups(p<0.01).Conclusion:1.TLR4 and TNF-?in rats infected with mycoplasma pneumonia increased.2.Zhiyan mixture and azithromycin can inhibit the concentration of TLR4 and TNF-?to some extent.3.Zhiyan Mixture can improve the inflammatory response of rats infected with mycoplasma pneumonia,and the mechanism may be related to the inhibition of TLR4 and TNF-?expression.
