Objective: To observe the effects of Zhiyan Mixture on the general conditions,pathological morphology of lung tissues,serum IL-6 and complement C3 of rats infected with Mycoplasma pneumoniae,and to analyze the changes of IL-6 and complement C3 in serum of rats in different groups,so as to explore the possible mechanism of Zhiyan Mixture in the treatment of rats infected with Mycoplasma pneumoniae.Methods: Forty-eight SPF rats,half male and half female,weighing about(200±20)g,were randomly divided into blank group,model group,azithromycin group and Zhiyan mixture group,with 12 rats in each group.Rat MP pneumonia model was established by nasal drip of MP strain for 3 days except for blank group.The blank group and model group were given normal saline intragastrically,and the azithromycin group was given azithromycin at a dose of 10mg/200 g on the first day and at a dose of 5mg/200 g on the fourth day.In the Zhiyan Mixture group,the rats were gavaged with Zhiyan Mixture(about 1ml/ time,twice/day)according to the body surface area of the rats,for 10 days.The rats were sacrificed 5 and 10 days after administration,respectively.The lung tissue samples were collected for HE staining and serum IL-6 and complement C3 content were measured by ELISA method to observe the therapeutic effect of Zhiyan mixture on MP-infected rats and the possible immunological mechanism of Zhiyan mixture and azithromycin in the treatment of MPP.Results:1.The results of HE staining showed that the alveolar wall of the lung tissue in the blank group was not widened and broken,and the structure was intact without injury.No congestion and edema were observed in the lung parenchyma and interstitium,and no obvious inflammatory cell infiltration was observed.In model group,lung interlobular septa and alveolar wall were widened,thickened and fractured.Interlobular vasodilation,alveolar congestion,interstitial edema with a large number of inflammatory cell infiltration;The pathological results of Zhiyan Mixture group and azithromycin group were between model group and blank group.2.The pathological score results showed that the pathological score of the other three groups was different from that of the blank group on the 5th and 10 th day after intervention(P<0.05);The pathological score of azithromycin and Zhiyan mixture group was also different from that of model group(P<0.05).There was no significant difference between azithromycin and Zhiyan mixture(P>0.05).At the 10 th day of treatment,compared with the 5th day after treatment,there was no statistical difference between the two groups before and after treatment(P>0.05).3.The results of IL-6 and C3 tests showed that :Serum IL-6 and complement C3 contents of the three groups inoculated with MP were increased during the whole experiment except blank group(P<0.05).Among them,the increase of serum IL-6 and complement C3 in the model group was the most obvious(P<0.05).The serum IL-6and complement C3 contents in the azithromycin group and the Zhiyan mixture group were both between the blank group and the model group on the 5th and 10 th days(P<0.05).Conclusion:1.Zhi Yan Mixture and azithromycin can relieve the symptoms of rats infected with Mycoplasma pneumoniae,and reduce the pathological structure damage,congestion and edema of lung tissue and inflammatory cell infiltration.2.Serum IL-6 and complement C3 increased in MP-infected rats.3.Zhiyan Mixture and azithromycin can reduce the concentrations of IL-6 and complement C3 in serum of MP-infected rats.4.Zhiyan Mixture can effectively reduce the inflammatory response of lung tissue,and the effect is better than that of azithromycin,which may be related to the fact that Zhiyan Mixture inhibits the expression of serum inflammatory factors and the activation of the complement system.
