Objective: to establish a mouse model of chronic nonbacterial prostatitis(CNP)and observe the effect of musk combined with mastic on substance P in prostate tissue.Methods: 48 male NOD/SCID mice aged 6 weeks were randomly divided into 4 groups: blank group,model group,musk-mastic group and musk-mastic high-dose group.CNP model was established in all mice except the blank group.Lavage in the 21 st day after the injection of drugs,the blank group,model group give the corresponding normal saline lavage,continuous lavage after 14 days to take blood,get off the neck executed after blood and anatomical isolate prostate tissue in mice,HE staining to observe the prostate gland inflammation,cell immunohistochemical observation the change of substance P in the prostate tissue,ELISA method to detect the variation in content of substance P in serum of mice.Results: 1.Through observation,we found that the glands in the prostate tissues of mice in the model group were dark red and generally adhered to the surrounding tissues,presenting pathological changes of chronic inflammation.After HE staining,microscopic observation showed that the pathological manifestations of the model mice were significantly different from those of the normal group,indicating that the model was successfully built.2.Immunohistochemical observation model group substance P positive cells was significantly increased in the blank group,the image analysis of optical density value,it is concluded that the average optical density value,the model group compared with the blank group of substance P in the prostate tissue increased obviously,statistically significant difference(P < 0.05),after lavage musk-frankincense,substance P decreased obviously in the prostate tissue in mice,statistically significant difference(P < 0.05),high dose-frankincense and musk-frankincense group comparison of substance P decline slightly,differences have no statistical significance(P > 0.05).3.ELISA showed that substance P content in the model group was obviously increased,compared with the blank between way differences statistically significant(P < 0.05),musk,frankincense group compared with model group statistically significant difference between the substance P decreased groups(P < 0.05),musk,frankincense,high dose group compared with musk,frankincense group substance P content is a slight decline,compare the differences between groups have no statistical significance(P > 0.05).Conclusion: 1.Musk-frankincense can reduce the content of substance P in the prostate tissue of CNP model mice,which indicates that musk with frankincense has the effect of "removing stasis and turbidity".Its "stasis" contains substance P,and can achieve the purpose of "removing blood stasis and clearing up" by reducing the release of substance P.2.Musk-mastic had no dose-dependent effect on the content of substance P in prostate tissues of CNP model mice.
