Objective:To investigate the expression changes of Wnt-5a and protein kinase C(PKC)in skeletal muscle during limb-ischemia-reperfusion injury(LI-RI)and the effect of pretreatment of peach four liquid solution on its expression Further,the molecular mechanism leads to the pathological mechanism of LI-RI and the therapeutic effect of Taohong Siwu liquid on LI-RI.Methods:78 SPF healthy male SD rats were randomly divided into normal control group,model group(M-IR group),Taohong Siwu liquid group(T-IR group)and phospholipase C blocker group(P-IR group).Four groups were included,in which the normal group did not cause ischemic treatment,and the remaining M-IR group,T-IR group and P-IR group were grouped at 0h,2h,4h,8h,etc.after recovery..After taking the materials,HE staining and real-time PCR were used to observe the morphological changes of rat gastrocnemius muscle and the expression of Wnt-5a and PKC in the gastrocnemius muscle.The data were statistically processed.Results:In addition to the normal group,the results of HE staining in M-IR group,T-IR group and P-IR group showed that the gastrocnemius muscle fibers were thickened and edematized in different degrees,and the interfiber space was obviously widened and structurally disordered.There are varying degrees of inflammatory exudation and apoptosis.Compared with the normal group,the mRNA expressions of Wnt-5a and PKC in the gastrocnemius muscle of M-IR group,T-IR group and P-IR group were increased to different degrees,and the difference between the groups was statistically significant(P< 0.05),and the relative expression levels of Wnt-5a and PKC mRNA in T-IR group and P-IR group were lower than those in M-IR group at the same time after reperfusion(P<0.05).Conclusion:1.The expressions of Wnt-5a and PKC in skeletal muscle of rats with limbischemia-reperfusion injury were increased to varying degrees,and skeletal muscle injury was severe,indicating that skeletal muscle tissue damage in LI-RI rats may be related to Wnt-5a and PKC.The high expression is closely related.2.Taohong Siwu liquid can reduce the high expression of Wnt-5a and PKC in skeletal muscle tissue of LI-RI rats,thus reducing the degree of LI-RI and protecting the skeletal muscle.
