ObjectiveAbserve gastrointestinal dynamics change of the functional dyspepsia rats,to discusses different acupuncture order to the difference of gastrointestinal regulation of rats,trying to explain the importance of the sequence of operation by compare the differences in the relevant data.MethodsThe experiment is divided into 1 and 2,experiment 1 randomly divided 12 rats into blank group and model group,6 rats in each group.The blank group was fed normally,and the model group was continuously molded for 21 days with mild clipping stimulation and intermittent diet,used to evaluate the decision FD model.A total of 60 rats in experiment 2,they were randomly divided into the blank control group,the model control group,the Upper and Middle Jiao group,the Middle and Upper Jiao group and the Middle and Lower Jiao group,with 12 rats in each group.Except the blank control group,all the other groups adopted the same molding method as the model group of the experiment 1.After successful modeling,each group was given corresponding intervention(there were 3 groups of acupoints for manual intervention,?Juque?Tiantu;?Lanmen?Jianli;?Qihai?Guanyuan.The manipulation intervention is carried out in the following order:the Upper and Middle Jiao group:we do ? and then do ?,while the Middle and Upper Jiao group:we do ? and then do ?,the Middle and Lower Jiao group:we do ? and then do?.Each acupoint was stimulated for 2min,a total of 8min.The blank control group and the model control group were fixed for 8min at the same time every day.Each group was continuously intervention for 14 days).The general mental activity status,behavioristics,electrogastric activity,gastric residual rate,intestinal movement rate and c-kit expression in gastric antrum of each group were recorded.The results of experiment 1:1.General observation:During the mold-making period,the rats in the blank group were in good mental state,their hair was smooth and shiny,and their weight steadily increased.In the model group,the hair of the rats was dry,yellow and lustrous,and there were behaviors and symptoms such as dim eyes,narrow eyes,mental tiredness and even clumping together.before model establishment there was no significant difference in weight between the two groups(P>0.05),and the weight of the model group was decreased compared with that of the blank group after model establishment(P<0.05).2.Pathological observation:In the model group and the blank group,the observation under HE staining was consistent:the gastric mucosa was intact,there was no ulcer or erosion,the structure was complete,and the gastric muscle layer was intact.The number of wall cells is normal.3.Gastric residual rate and intestinal movement rateThe intestinal propulsion rate of the model group was lower than that of the blank group(P<0.05).The results of experiment 2:1.General observationAfter manipulation intervention,the mental state of each group of rats was improved compared with the previous group.The hair gradually recovered smooth and bright,the color around the eyes gradually weakened,and the fecal character gradually returned to normal.model control group rats's general status at the end of the building is slightly better,but with occasional cluster phenomenon,and not easily controlled when fetching fixed,compared with the blank control group,rats state of mental activities still has bigger difference.2.Behavioral changeAfter manipulation intervention,the number of horizontal and vertical movements of rats in the model control group was lower than that of the blank control group(P<0.05).3.electrogastrogramCompared with the blank control group,the EGG amplitudes of the model control group was lower than that of the blank control group(P<0.05).Compared with the blank control group,the EGG amplitudes of the Upper and Middle Jiao group and the Middle and Lower Jiao group were no statistically significant(P>0.05).Compared with the model control group,the EGG amplitudes of both the Upper and Middle Jiao group and the Middle and Lower Jiao group were higher than those of the model control group(P<0.05).4.Gastric residual rate and intestinal movement rateCompared with the blank control group,the gastric residual rate of the model control group was higher than that of the blank control group(P<0.05),and the intestinal movement rate was lower than that of the blank control group(P<0.05).Compared with the blank control group,the gastric residual rate and intestinal movement rate in the the Middle and Lower Jiao group were no statistically significant(P>0.05).Compared with the model control group,the gastric residual rate in the Middle and Lower Jiao group was lower than that in the model control group(P<0.05),and the intestinal movement rate was higher than that in the model control group(P<0.05).Compared with the control group,the small intestine movement rate in the Middle and Lower Jiao group was higher than that in the Upper and Middle Jiao group and the Middle and Upper Jiao group(P<0.05).5.Changes of c-kit expression in gastric antrumCompared with the blank control group,the mean optical density(OD)of c-kit in the model control group was lower than that of the blank control group(P<0.01),and the positive ICC of c-kit was decreased and the color was lighter.There was no statistically significant difference between the OD of the manipulation intervention group and the blank control group(P>0.05).Compared with the model control group,the OD of the Middle and Lower Jiao group was higher than that of the model control group(P<0.05),and the positive ICC of c-kit increased and the color became darker.Conclusion1.In experiment 1,combined with emotional factors to making model,the rats in the model group showed symptoms such as loss of appetite,yellow hair and lack of luster,and no organic lesions were detected by visual and pathological examination.Weight loss in the blank group and small intestine movement rate in the blank group were lower than that in the blank group,indicating that the symptoms were similar to those in the pathology and FD patients,which could prove that the animal model of FD was successfully reproduced in this experiment.2.Zang-fu viscera acupoint pressure therapy can effectively improve the activity state,cognition and exploration ability of FD rats,and can alleviate the depression,loss of interest,mental depression and motor retardation of rats.In addition,the EGG amplitudes of FD rats were increased to improve the disorder of gastric electrical rhythms and to regularize the movement of the stomach.It can also promote gastric emptying and intestinal propulsion,and improve gastrointestinal motility in different order of acupoint.3.The effect mechanism of zang-fu viscera acupoint pressure therapy may be increase the expression of c-kit,change the number of ICC and structure,mature and regulating gastric electric rhythm,improve rats gastrointestinal disorders,which enhance the contraction of gastric antrum,promote gastric emptying.4.In the order of acupoint treatment for FD,the best curative group is the Middle and Lower Jiao group.first point the Middle Jiao and then point the Lower Jiao is more effective than first point the Middle Jiao and then point the Upper Jiao.
