Experimental Study On The Effect Of Jianwei Qutong Recipe On Hp-related Chronic Non-atrophic Gastritis In Rats

Objective:To observe the effects of Jianwei Qutong Recipe on the levels of TNF-?,IL-2 and IL-4 in rats with Hp-related chronic non-atrophic gastritis,and to explore the possible mechanism of of Jianwei Qutong Recipe in the treatment of Hp-related CNAG from the perspective of inflammatory factors,which provide a certain experimental basis for clinical applications.Methods:48 SD male rats were randomly divided into normal group?10rats?and model group?38 rats?.Amoxicillin solution+NaHCO₃+Hp SS1bacterial solution A rat model of Hp-related chronic non-atrophic gastritis was established by a method of intragastric administration.Successful modeling were randomly divided into 3 groups according to body weight,10 in each group,including 4 groups in the original normal group,namely normal group,model group,Jianwei group and quadruple grop.The normal group was fed with sterile water;The model group was fed with sterilized water;The Jianwei group and the quadruple group were given different drugs for gavage.and the rats with Hp-related chronic non-atrophic gastritis were observed after 14 days of continuous intervention.The effects of serum TNF-?,IL-2,IL-4,urease content in gastric mucosa,and histopathological changes of gastric mucosa.Result:1.At the 5th week of moulding,HE staining of gastric tissue in the model group showed disorder of gastric mucosal epithelial cells,and more lymphocytes and neutrophils infiltrated into the mucosa.The results of gastric tissue examination showed that the Hp-related CNAG rat model was successfully replicated.2.After the end of the treatment,compared with the model group,the IL-4content in Jianwei group was significantly increased,the difference was statistically significant?P<0.05?,and the contents of TNF-?,IL-2 and urease did not change significantly.3.After the end of the treatment,Compared with the normal group,the contents of TNF-?and IL-2,IL-4 in the model group,Jianwei group and quadruple group were significantly higher than those in the normal group?P<0.05?.compared with the model group,HE staining of gastric tissues in the jianwei group and the quadruple group showed less changes in mucosal inflammation;Inflammation score showed a downward trend,but there was no significant statistical significance.Conclusion:1.Rat model of Hp-related chronic non-atrophic gastritis can be successfully established by intragastric administration of Amoxicillin solution+NaHCO₃+Hp solution.2.Compared with the 4th week,the degree of gastric mucosal inflammation and Hp infection was more obvious at 5th week,suggesting that modelling time could be prolonged appropriately to improve the success rate of modelling of Hp-related chronic non-atrophic gastritis in rats.3.Jianwei Qutong Recipe can significantly increase the content of anti-inflammatoryfactors IL-4 in serum of rat model of chronic non-atrophic gastritis associated with Hp,so it is speculated that it may reduce gastric mucosal inflammation by increasing the expression of anti-inflammatoryfactors IL-4.
