Surface-enhanced Raman scattering(SERS)detection is widely used in biomedical analysis due to its high sensitivity,specificity and non-destructive property.At the same time,the microfluidic chip has the advantages of miniaturization,integration,low sample cost,convenient operation and high reaction rate.This microfluidic chip has shown important application value in biological detection.Herein,a class of SERS microfluidic chips based on ordered array substrate is proposed,which improves the sensitivity and reliability of detection,and opens up a new way for point-of-care detection.The main work is as follows:First,an ordered nano-array silver substrate was prepared as a SERS substrate with an AAO template,which significantly improved the SERS enhancement factor and the uniformity of SERS detection.Using the mouse antigen as the analyte,a protein immunoassay with a detection limit of less than 0.01 ?g/mL was achieved.Secondly,SERS probes with various structures were prepared based on spherical gold nanoparticles.The SERS activity of probe was studied.The results show that the coating of silica shell has no obvious effect,but the existence of silver shell is beneficial to enhance the SERS intensity and helpful for fixation of Raman reporters.HeLa cell was recognized by a gold silver core-shell SERS probe modified with aptamers.Based on the above,a SERS-microfluidic multivariate detection platform based on ordered array substrate and gold silver core-shell SERS probe is proposed.The phenotypic analysis of leukemia cells can be realized with the whole blood sample of patients.The experimental results show that the ordered array substrate has higher cell capture efficiency(greater than 81%)and capture purity than the planar substrate.Finally,the SERS spectra of leukemia patients were used to train a linear SVM model.The results show that the model has a classification accuracy of 98.6%.The platform may have broad application prospects in leukemia phenotype analysis.
