Because of super sensitivity, high selection and soft detecting condition, SERS has been becoming to one of the important ways for proteins research. With the fast development of Surface-Enhanced Raman Scattering (SERS) technique, it is widely used in the field of bioscience research, especially in the conformational structure for proteins. This paper builds a PDMS-microfluidic chip with SERS activity to detect analytes. The main contents of this thesis include followings:SERS-active substrates on PMDS are made and its performances are studied by chemical self-assembled method and liquid-liquid interface self-assembled method.To fabricate SERS substrate with chemical self-assembled method, the oxygen plasma treatment was used to PDMS surface modification, and then silver nanoparticles are assembled on the PDMS through electrostatic interaction or chemical bond linker. The substrates have good SERS activity and detecting sensitivity. The substrates by chemical bond linker method have better SERS activity because of some SERS 'hot pots'. SERS substrates with liquid-liquid interface assembled mental film are fabricated. In the interface of silver colloid and toluene, adding ethanol is to make the nanoparticles run to the interface quickly and to produce large silver film. Then the film is shifted to the PDMS slab by floating or pulling method to form compact, uniform and small- gap nanoparticles film. The pulling substrates with better SERS activity are for double structures.PDMS microfluidic chips are fabricated and the SERS signals of analytes are detected online. Through method of molding, the PDMS cover slab with micro-channel and substrate slab with SERS activity is made. SERS chips are produced by bonding the two one.1.0 X 10-6M R6G liquor is used as SERS probes to help analyze the performances of chips,and the chips are better than solid substrates. At last, the SERS for proteins on the surface of open substrates and in the chip is detected.
