As the most revolutionary new area of clinical medicine, Point-of-Care Testing technology has advanced a lot in clinical applications because of its small volume, portability, easy-using and rapid results. As the application goes much wider and deeper, there is a need for miniaturizing and integrating more and more analytical devices to reduce it to lower cost, more convenient, smaller and faster way to provide the information of material ingredient and structure.μTAS is the technology to integrate the analytical lab into portable device and finger size chip. This dissertation describes the microfluidic immunoassay based on the clinical immunochemical technology in the background of above.The topics of this dissertation are presented below:1. After the in-depth research of the backgrounds, characteristics, applications and tendency of POCT technology, solutions of Microfluidic immunoassay are presented to integrate the immunochemical technology.2.The quality of SU-8 mold has been improved along the process of microfluidic chips, and the adhesion between mold material SU-8 and Si substrate, thus the process solution has been optimized. The mold is able to transfer pattern to PDMS microfluidic chip for many times, therefore reduces cost.3. The surface modification and antibody immobilization technology are studied along with the immobilizing efficiency of different immobilizing carrier and different material based on PDMS microfluidic chip, and then the way of immunochemical analysis has been successfully built.4.The fluid mixture process has been studied under power-free driven technology through designs of chip structure and numerical analysis. Then the fluidity, fluid velocity and reaction sufficiency of immunoreaction has been studied based on this chip platform. The results have been validated and the sensitivity has been effectively improved.It has been shown that the SU-8 microfluidic mold has the advantages of being reused for many times, low cost, easy for fabrication etc. The PDMS microfludic immunoassay could detect parallel in the way of multi-channel and improve the detection sensitivity. It is the key part of POCT systematic of microfluidic immunoassay.
