Benefiting from the rapid development of microelectronics technology and micro-electro-mechanical technology,microfluidics has been widely used in the fields of biology and environment.In the field of microfluidics,China starts late and has been lagging behind the United States and Europe,especially in the field of microfluidic analysis and detection instrument.In terms of function,there is a large gap between China's self-developed products and foreign products.As a result,it is of great strategic importance to thoroughly study the microfluidic analysis and detection instrument.In this thesis,a set of miniaturization,high integration,and low detection limit micro-droplet optical chip fluorescence detection platform is designed and constructed.The dynamic concentration detection range of the platform is 5n M-100 n M.The platform's linear correlation is 0.9962,the minimum detection limit of small molecule mi RNA is 5n M.Firstly,the micro-droplet optical chip is designed and manufactured by micro-electro-mechanical technology,it includes a micro-droplet generation module,an excitation fiber insertion port,and a micro-lens.The micro-droplet chip is replaced by a micro-droplet chip integrated with a micro-lens,which improves the integration of the device and enhances the collected fluorescence intensity and reduces detection limit.Secondly,the micro-droplet optical chip fluorescence detection platform is used to mi RNA detection,the molecular probe system in which micro-droplet-encapsulating mi RNA is excited by light,mi RNA and molecular probe combine to generate fluorescence resonance energy transfer,signals of Cy3 and Cy5 are detected by two PMTS.In addition,instead of the traditional objective lens excitation light path,optical fiber has reduced the volume of the entire space and achieved the miniaturization of the fluorescence detection platform.The use of micro-droplet has reduced the amount of detection objects,and solved the ribozyme treats degradation of the test substance in the experimental environment.
