Fluorescence microarray chips and their corresponding detection methods is a high and new technology,which is developing rapidly in recent years by our nation's strong support.It has some attracting features including high detection efficiency and accuracy,which causes a revolution in life science research,disease diagnosis,new drug research and development,forensic authentication,food hygiene supervision,etc.At present,foreign research on the development of microarray chip detection device has a certain result,some company have put it into production.Compared with foreign countries in the development of the detection equipment,it is relatively slow and relatively small in our country,resulting in the domestic equipment is not only complex and high cost of the device.Therefore,a CCD-based fluorescence microarray chip detection device is designed and developed,which simplifies the structure of the detection device and improves the detection speed while reducing the cost,according to the actual situation in terms of image processing for chip of irregular lattice,"four pyramid" model is used in grid processing.In this paper,the research content can be divided into the following several aspects:(1)We introduce fluorescent microarray chip technology and fluorescence detection principle of micro point on the chip,then illustrate the fluorescence acquisition process of detection device and two common way of microarray chip test.(2)According to the fluorescent properties of fluorescent marker dye,choosing the appropriate excitation light source and optical devices.Then designing the optical engine with light path system.At the same time,designing STM3 2?temperature control and stepper motor hardware circuit.RTX real-time operating system based on KEIL is also used to provide the Collaborative service for the detection device.Finally the standard curve of the CCD response is examined.(3)Aiming at the irregularity of the microarray on the chip in the actual process,the"quadrangular pyramid" model is adopted in the step of the image processing to realize the problem of microarray irregularity,and the grid result of the method is obviously better than the projection method,which provides a guarantee for the next step of data processing.(4)At the end of this paper,the detection sensitivity,resolution and repeatability of the detection device are tested and analyzed,and the detection of genetic defects in Beijing Taipu has been achieved,the original image and the fluorescence gray scale on the chip has been obtained,which lays the foundation for the subsequent biochemical analysis,while discussing the problem and improvement of the device.The experimental results show that the detection sensitivity of the fluorescence microarray chip detection device designed and developed in this paper is 0.01?mol/L,Its repeatability error within 4%,resolution reached 10.05pm,the noise performance of the device is stable,the fitting of the standard curve finally finds the fitting of the cubic spline,which can reduce the detection error to the maximum extent.
