Objective: Adipocyte differentiation and thermogenesis play an important role in the energy expenditure of adipose tissue.BMP-4 and BMP-7 are important regulators of the differentiation of stem cells into adipocyte lines.They also have important regulatory roles in the thermogenesis of mature adipocytes.Therefore,this study will be carried out through moderate intensity exercise and high-intensity intermittent exercise for obese rats,to observe the differentiation and thermogenesis of adipocytes in straight tissue.It provides a theoretical basis for the mechanism of exercise promoting the energy consumption of adipose tissue,and provides guidance for the scientific fitness of obese people.Methods:80 3-week-old male SPF SD rats were fed with high-fat-diet for 8 weeks,30 11-week-old obese rats were obtained from these rats.The 30 obese rats were randomly divided into 3 groups: sedentary obese group(SO,n=10),moderate-intensity endurance obese group(MO,n=10)and high-intensity interval obese group(HO,n=10).20 11-week-old chow-diet rats were randomly divided into sedentary chow-diet group(SC,n=10)and moderate-intensity endurance chow-diet group(MC,n=10).The rats in each group remained the same diet habit,rats in the MC and MO attended moderate-intensity endurance training(60%-70%V O2 max,50 minutes/day,5 days/week)and HO attended high-intensity interval training(Running alternately with 50% V O2max?70% V O2max?90% V O2 max,50 minutes/day,5 days/week)for 8-weeks.At the end of the 8 week,abdominal aorta,scapular brown fat,subcutaneous white adipose tissue and periepididymal white adipose tissue were taken from all rats.All rats were fed daily food intake,weighed and body length were measured weekly,and the protein expression levels of BMP-4,BMP-7,TBX1,PRDM16 and UCP-1 in adipose tissue were detected by Western blot.Result:(1)Exercise significantly reduced the body weight and the Lee's index of obese rats,and also reduced the levels of serum total cholesterol and triglyceride in obese rats(P<0.05).The effect of HIIT is stronger than that of moderate intensity exercise on the above indexes.(2)The increase in the expression of UCP-1 protein appearsed in in the brown adipose tissue and subcutaneous adipose tissue of obese rats(P<0.01).(3)Mmoderate intensity exercise significantly increased the expression of BMP-4 TBX1 and UCP-1 protein in subcutaneous fat of obese rats(P<0.01);HIIT significantly promoted the expression of BMP-4 TBX1 and UCP-1 protein in peri-testis fat(P<0.01);Moderate intensity exercise and HIIT could inhibit the expression of BMP-7 and UCP-1 protein in brown fat(P<0.05),and moderate intensity exercise could inhibit the expression of BMP-7 and UCP-1 protein more strongly.Conclusions: Adipose tissue of obese rats,especially brown adipose tissue,has a certain degree of increase of calorific metabolization-associated proteins.Exercise can promote the expression of BMP-4 protein and calorific metabolization-related protein in White adipose tissue;And exercise reduced the expression of BMP-7 protein of the brown adipose tissue and its thermogenesis.The effect of different sports methods on different tissues is different.Whether the changes of exercise induced BMP-4 and BMP-7 are involved in the regulation of the calorific metabolism of adipose tissue and the regulatory mechanism need to be further studied.
