The Influence Of Dectin-1 On IL-6 And IL-17 In The Incubation Of Spleen Lymphocytes And Heat-treated Candida Glabrata

Objective: In the intensive care unit(ICU),the threat of invasive candidiasis caused by Candida glabrata is growing rapidly.To explore the role of dendritic cell-associated C-type lectin-1 protein(Dectin-1),interleukin-17(IL-17)and interleukin-6(IL-6)in the immune process induced by heat-treated Candida glabrata by establishing MSLs incubation model with heat-treated Candida glabrata.Methods: MSLs were randomly divided into four groups.Lymphocyte stimulant concanavalin A was added in negative control group(Group B),fungal incubated group(Group C)and pre-inhibited incubated group(Group D),while the blank control group(Group A)was added with the same amount of PBS.Laminarin,a specific inhibitor of Dectin-1,was added in group D.Group C and group D were incubated with heat-treated Candida glabrata for 0,2,4 and 6 hours,respectively.The morphological changes of cells at each time point were observed under light microscope,the survival rate of MSLs at each time point was measured by CCK-8 method,and the concentrations of IL-17 and IL-6 in the culture medium supernatant at each time point were measured by ELISA method.Results: Incubation with heat-treated Candida glabrata could cause MSLs injury and aggregation,but the injury was more serious and the aggregation was less in group D,and the expression of IL-17 and IL-6 in group C and group D were both higher than that in group B at each time point(P<0.05),and the concentration of IL-17 and IL-6 in group C was higher than that in group D after the incubation(P<0.05).Conclusions: Dectin-1 plays a certain protective role in the process of heat-treated Candida glabrata incubated with MSLs.The recruitment of immune cells in this process may be mediated by Dectin-1 up-regulating the expression of IL-6 and promoting the secretion of IL-17,by Th17 cells,and the cross-talk of innate immunity and acquired immune response through IL-17.
