Role Of Dectin-1 Protein,IL-17 And IL-23 Levels In Rats With Invasive Heat-treaed Candida Glabrata Infection

Objective: To investigate the changes and significance of dendritic cell-associated lectin-1(Dectin-1)along with levels of interleukin-17(IL-17)and interleukin-23(IL-23)in serum and broncho alveolar lavage fluid(BALF)of rats with invasive heat-treated Candida glabrata infection.Methods: Fifty-four SD rats were randomly divided into control group(group A),direct infection group(group B)and immunosuppressive infection group(group C).Each group had 18 rats.No treatment applied on Group A while group B was perfused with heat-treated Candida glabrata through trachea directly.At the same time,rats in group C were perfused with equal dose of heat-treated Candida glabrata after immunosuppressive treatment.The pathological changes of lung tissue were observed on the 1st,3rd and 5th day after infection,the expression of IL-17 and IL-23 in alveolar lavage fluid and serum were detected by enzyme-linked immunosorbent assay(ELISA),and the expression of Dectin-1 protein in lung tissue was detected by Western Blot.Results: Lung inflammation and lung injury were the most severe in group C,and there was no inflammation in group B after group B.The levels of IL-17 and IL-23 in serum and alveolar lavage fluid and the expression of Dectin-1 protein in lung tissue in group A were not significantly different at all time points.In group B and group C,the levels of IL-17 and IL-23 and the expression of Dectin-1 protein in lung tissue increased with time,while the levels of IL-17,IL-23 and Dectin-1 in group C were significantly higher than those in group A and group B at each time point.Based on Multiple Regression Analysis,the levels of IL-17 and IL-23 as well as Dectin-1 protein were impacted by different types and various time length of immunosuppression and fungal infection on lab rats.The most noticeable effect is observed from immunosuppression to IL-17,IL-23 and Dectin-1 protein Dectin-1 while the infection and time control of fungal infection follows.Conclusion: Dectin-1 is a critical receptor for identification of heat-treated Candida smooth infection.Induced IL-17 and IL-23 participate in antifungal immune response and may cause lung injury.
