Objective: To investigate the effect of Dectin-1 on the release of inflammatory factors interleukin-10(IL-10)and tumor necrosis factor-?(TNF-?)in rats tracheal epithelial cells(RTECs)stimulated with heat-treated Candida glabrata.Methods: RTECs in vitro were divided into three groups,such as the control group,the heated Candida glabrata stimulation group and the receptor block group.Then we harvested the cells at 0,2,4,6 hours.The cell survival rate was determined by MTT.Futhermore,the expressions of cytokines IL-10 and TNF-? were analyzed by ELISA.Moreever,the protein expression of Dectin-1 were analyzed by Western-blot.Results: Our data indicated that heat-treated Candida glabrata contribute to the inhibition of RTECs.The expressions of Dectin-1,IL-10 and TNF-? were increased after stimulated with heat-treated Candida glabrata(p<0.05).After pretreated with Laminarin,the expressions of Dectin-1,IL-10 and TNF-? would all decreased compared with the other two groups at the same point(p <0.05).Conclusion: Dectin-1 plays an important role in recognizing the heat-treated Candida glabrata of RTECs,and it leads to the immune responses and cytokines production,for instance,IL-10 and TNF-?.Laminarin plays an inhibitory effect in the production of IL-10 and TNF-? by RTECs stimulated with heat-treated Candida glabrata.
