| Objective : Observe the therapeutic effect of Mahuang Lianqiao Chixiaodou Decoction in the clinical and animal experiments,give a more comprehensive evaluation.We hope explore the new uses of the traditional Chinese medicine,provide a scientific basis for modern innovative applications of Mahuang Lianqiao Chixiaodou Decoction also find out a new treatment method for Ig AN.Methods: Part1: 53 patients with dampness-heat syndrome of Ig AN were randomly divided into treatment group and control group by using the method of random single blind and parallel design.The patients were treated with Mahuang Lianqiao Chixiaodou Decoction and ACEI/ARB respectively.After 8 weeks of treatment,the changes of clinical symptoms,urine red blood cell count,24-hour urinary protein quantity,serum creatinine,urea nitrogen,plasma albumin and immunoglobulin were observed in the two groups before and after treatment.At the same time,the changes of safety indexes such as liver function were observed.Part2: In this study,60 male SD rats were randomly divided into normal group(12)and model group(48)this two groups.Ig AN rat model was established by taking BSA solution alternate days for 8 weeks,subcutaneous injection CCL4 weekly for nine week,and injecting LPS into tail vein at sixth week and eighth week.When forming the model,rats were randomly divided into 4 groups according to their 24-hour urinary protein level,12 rats in each group: model group,Mahuang Lianqiao Chixiaodou Decoction group,rituximab group and losartan potassium group.After 8 weeks of treatment,physiclal status,urine and serum indexes were measured at the14 th and 18 th week of the experiment,the fluorescent expression of Ig A,and the pathological staining of the kidneys were detected too.Results: Part1: The total score of the treatment group was significantly lower than that of the control group before and after treatment(p < 0.05),and the improvement of clinical symptoms was better than that of the control group(p < 0.05).The total effective rate of the treatment group was 53.72%,which was statistically better than that of the control group(p < 0.05),effectively reduce albuminuria,hematuria,protect renal function,regulate humoral immunity.Part2: After modeling,the rats were torpescence,and their hair clutter,weight loss,proteinuria in(p < 0.001),and obvious deposition of Ig A immune complex in glomerular mesangial area(p < 0.05).After 8 weeks of treatment,Mahuang Lianqiao Chixiaodou Decoction significantly improved the physiclal status and reduced the mortality of rats(p < 0.05).Compared with the model group,the albumin uria decreased(p < 0.05),and the curative effect was the best.The serum Scr decreased significantly(p < 0.05),and the intensity of fluorescence deposition in mesangial area decreased(p < 0.05),which indicated that Mahuang Lianqiao Chixiaodou Decoction was effective in treating Ig AN.Conclusions: In the clinical and animal experiments of treating Ig AN,Mahuang Lianqiao Chixiaodou Decoction has a excact therapeutic effect. |
