Modern research shows that liquor is a liquid composed of a variety of chemicals,of which about 98% are water and ethanol,and the remaining 1%~2% are some trace organic compounds produced during solid-state fermentation.The organic composition ratio of this part forms the unique flavor and style of liquor,which directly affects the flavor and taste of wine.Secondly,after the interaction of various organic substances in liquor,new effective substances and functions are produced,and the comprehensive effects on human body become more complex.According to the report of the World Health Organization,there are about 3 million people die of alcohol abuse every year,which has become the third largest risk factor for the global burden of disease.With the increase of alcohol-addicted population in China,the harm of alcohol to various organs of human body,especially the metabolic organs such as liver and kidney,is becoming more and more obvious.In the 21 st century,with the deepening of consumers perceptions of healthy drinking,researchers have more and more studies on the relationship between drinking and health,and gradually turned the past research focusing on phenotype to the mechanism Mining of body damage and the protection mechanism of trace components in drinking alcohol.Therefore,this topic was proposed to study the toxic effect of alcohol exposure on liver and kidney tissues in mice and its molecular mechanism.(1)In this experiment,C57BL/6J mice were used to establish a gavage model,and the effects of drinking alcohol and eating alcohol on liver and kidney tissues of mice were compared.The experimental results showed that the liver and kidney of mice showed a certain degree of damage effect after drinking and eating alcohol exposure,the levels of reactive oxygen radical ROS and lipid peroxidation product MDA were up-regulated,and the content of superoxide dismutase SOD was reduced,indicating the occurrence of oxidative damage;oxidative stress reaction triggered the initiation of autophagic reaction,resulting in a large number of autophagosomes and lysosomes.The most direct reflection is the aggregation and high expression of LC3 B and beclin-1,digestion and degradation of cells in vivo,thus causing death of hepatocytes,leading to a large expression of tumor suppressor gene P53,and the ratio of pro-apoptotic factor Bax/anti-apoptotic factor Bcl-2 is up-regulated.In addition,we also found that the degree of body damage caused by drinking alcohol exposure was less than that caused by eating alcohol,suggesting that some trace components in drinking alcohol may be responsible for this phenomenon.(2)In order to further explore the effect of micro-organic components in liquor on liver injury in mice,this study used high-acid and high-ester liquor samples for gavage model exposure,and detected the damage effect of liver tissue in mice.Corresponding experimental results showed that when the content of acetic acid,ethyl acetate,lactic acid and ethyl lactate in drinking alcohol was doubled,it could effectively slow down oxidative stress response and the occurrence of cell autophagy and apoptosis,alleviate the liver injury of mice caused by drinking alcohol exposure,and prove that some trace components in drinking alcohol could prevent alcoholic liver injury.
