Objective:To explore the effect of colonic dialysis combined with Yishen decoction on serum autophagy-related proteins Beclin1 and LC3? in CKD 3-5 stage patients.The purpose of this study is to provide a new way to find the intervention target of CKD.Methods:From March 2018 to July 2019,a total of 185 cases of CKD patients whose data was complete in our hoptital were collected.Then according to the inclusion and the exclusion criteria,finally 96 CKD 3-5 stage patients were selected as the CKD group,and 20 healthy volunteers from the physical examination centre were selected as the healthy group.Then the patients in the CKD group were randomly divided into control group(A group),colonic dialysis group(B group),and the colonic dialysis combined with Yishen decoction group(C group),there was 32 CKD patients in every group.The ones in the control group were given routine symptomatic treatment,the ones in the colonic dialysis group were added colonic dialysis based on the treatment of the control group,while the ones in the colonic dialysis combined with Yishen decoction group were added coloclysis of Yishen decoction based on the treatment of the colonic dialysis group.Colonic dialysis was performed 3times a week,2 hours a time,and the observation period was 4 weeks.Collected healthy volunteers' s general information,laboratory indexes,and the general information,laboratory indexes before and after treatment of CKD patients,meanwhile used enzyme linked immunosorbent assay(ELISA)to detect the levels of serum Beclin1,LC3? in the healthy volunteers and the CKD patients before and after treatment.The comparison of the general information,laboratory indexes between CKD group and healthy group was conducted,and the comparison of general information,laboratory indexes between the control group,the colonic dialysis group and the colonic dialysis combined with Yishen decoction groupbefore and after treatment was also conducted.Statistical method was used to analyze the effect of colonic dialysis combined with Yishen decoction.Results:(1)The the levels of serum Beclin1,LC3? in CKD 3-5 stage patients of CKD group were lower than the healthy volunteers of healthy group(P < 0.05),the difference was statistically significant.(2)There was no statistically difference in the levels of Scr,BUN,UA,ALB,TG,TC,HDL,LDL,Ca,P,PTH,Hb,hs CRP,Beclin1,LC3? among CKD patients of the three groups before treatment(P>0.05).(3)Comparison among the three groups before and after treatment : the levels of Scr,BUN,UA of the CKD patients in control group were lower than those before treatment(P<0.05),but the difference of the levels of Beclin1,LC3? before and afteer treatment had no significant difference(P>0.05);The levels of serum Scr,BUN,UA and hs CRP of the CKD patients in the colonic dialysis group and the colonic dialysis combined with Yishen decoction group were lower than those before treatment(P < 0.05),the levels of serum Beclin1 and LC3? of the patients in colonic dialysis group and colonic dialysis combined with Yishen decoction group were higher than those before treatment(P<0.05).(4)Compared with the control group,the levels of serum Scr,BUN,UA and hs CRP in CKD patients of colonic dialysis group and colonic dialysis combined with Yishen decoction group were reduced after treatment(P<0.05),and the levels of serum Beclin1,LC3? in patients of colon dialysis group and colonic dialysis combined with Yishen decoction group increased after treatment(P<0.05);Compared with the colonic dialysis group,the levels of serum Scr,BUN,UA,hs CRP in the CKD patients of colonic dialysis combined with Yishen decoction group were decreased significantly after treatment(P<0.05),the levels of serum Beclin1 and LC3? in patients of colonic dialysis combined with Yishen decoction group increased after treatment(P<0.05).(5)Correlation analysis:the levels of hs CRP(r=-0.425,P=<0.001)and Scr(r=-0.236,P=0.021)in CKD 3-5 stage patients before treatment were positively correlated w ith Beclin1,hs CRP(r=-0.404,P=<0.001)and Scr(r=-0.220,P=0.031)were positively correlated with LC3?.Conclusion:Colonic dialysis combined with Yishen decoction increased the levels of serum autophagy-related proteins Beclin1,LC3? in CKD3-5 stage patients,enhanced autophagy activity,improved the state of microinflammation,and delayed the progression of CKD.
