Objective: To establish a rat overactive bladder model,conduct animal behavioral tests,determine whether there is anxiety / depression and other emotional disorders in the model group,and verify the relationship between overactive bladder and emotional disorders in rats;energy metabolism in frontal lobe Western blot analysis whether there are differential expressions,to further explore the mechanisms related to overactive bladder and mood disorders.Methods: 1.The model of overactive bladder of rats was established by intraperitoneal injection of cyclophosphamide.2.Conduct rat behavioral experiments on rats at 2 weeks and 5 weeks after modeling to detect when rats have mood disorders.3.Differential expression of energy metabolism proteins GLO1 and CKB.Results: 1.Animal behavior experiments showed that the model of 2 weeks group had depression-like / anxiety-like behavior(P >0.05)2.After 5 weeks of modeling,the sugar preference of the model group and the control group were 68.49 12.73% and 81.05 9.63%(P <0.05)respectively.The model group 's preference for sugar water decreased and there was depression.3.In the elevated cross maze experiment after 5 weeks of modeling,the residence time of the model group and the control group in the open arm were 11.48 10.77 s and 60.86 24.54s(P <0.05),while in the closed arm the residence time was 244.88 32.22 s and 167.60 32.74s(P <0.05),the stay time of the model group in the closed arm is greater than the control group,and the stay time in the open arm is significantly shorter than the control group,indicating that the model group is more willing to stay in the safe closed arm,Anxiety-like behavior in rats;4.After 5 weeks of modeling,the rats in the model group compared with the control rats,in the open mine experiment,the central area stay for a long time,and the rats had anxiety(the central stay time is 10.54 4.44 s,4.91 3.44 s,P< 0.05);5.In the forced swimming experiment of rats,the still time of the 5 weeks model group in water was significantly longer than that of the control group(model group: 226.53 29.41s;control group: 186.33 26.29 s,P <0.05),and the bladder overdose can be obtained from the above behavioral tests The combination of active disorder model and mood disorders,especially anxiety / depression,and statistically significant difference with the control group(P <0.05);6.Western blot analysis showed that the GLO1 density value of the control group was lower than the model group(P> 0.05);the CKB optical density value of the model group was higher than the control group(P <0.05).Conclusion: Rats with overactive bladder may be associated with mood disorders in 5 weeks.And it is related to the energy metabolism of the frontal lobe of the rat cortex.Rats with overactive bladder may cause depression / anxiety due to increased CKB in the frontal lobe of the cerebral cortex,and depressed rats may also cause overactive bladder due to increased CKB in the frontal lobe of the cerebral cortex.
