Objective To investigate the single nucleotide polymorphism(SNP)rs2333227 of myeloperoxidase(MPO)gene of Han children in Inner Mongolia and its susceptibility to Kawasaki disease and coronary artery lesions(CAL)The relevance of this study provides some clinical data for the etiology of Kawasaki disease.Methods A case-control study was adopted,and 50 cases of Kawasaki disease children admitted to the Children's Hospital of Inner Mongolia Autonomous Region from October 2017 to January 2019 were selected as the case group.During the same period,50 children from the Infant Children's Hospital of Inner Mongolia Autonomous Region were selected as the control group.According to the presence or absence of coronary arteries,the cases were divided into CAL group and NCAL group..All research subjects used a collection tube containing Ethylene Diamine Tetraacetic Acid(EDTA)to draw 2 ml of venous blood,and DNA was extracted from the collected blood specimens using a DNA extraction kit.Polymerase chain reaction was used.,PCR)and direct sequencing of genes were used to detect the MPO gene SNP rs2333227 loci in the study subjects.SPSS 20.0 software was used to analyze the polymorphism of the MPO gene SNP rs2333227 loci in the case group and control group,CAL group and NCAL group.Come to the corresponding conclusion.Result All the research subjects detected the SNP at rs2333227 locus of MPO gene,and there were three genotypes of AA,GA and GG at this locus.The genotypes in this study conform to the Hardy-Weinberg equilibrium(P> 0.05)and are representative of the population: 1.AA,GA and GG genotypes of the rs2333227 locus of the MPO gene between the Kawasaki disease case group and the control group The frequency distribution was not significantly different(2 = 2.392,P =0.302).The frequency of the GG genotype was significantly higher than that of GA and AA in the case group and the control group.Compared with the GG genotype,the GA and AA genotypes were significantly different from the KD genotypes.The risk was not related(OR: 0.64,95% CI: 0.25-1.63).2.There was no significant difference in the frequency distribution of alleles(A and G)of rs2333227 locus of MPO gene between the KD case group and the control group(2 = 1.592,P = 0.351).3.The frequency distributions of the MPO gene rs2333227 loci AA,GA and GG were significantly different between the CAL group and the NCAL group(2 = 6.263,P =0.044),and the allele frequency distributions were significantly different(2 = 4.037,P= 0.045)Compared with GA and AA genotypes,GG genotypes increased the risk of CAL(OR: 5.13,95% CI: 1.22-21.63).4.Analysis of the correlation between the MPO gene rs2333227 polymorphism and clinical features of Kawasaki disease showed that rash,lip and oral manifestations,hand and foot symptoms,bulbar conjunctival hyperemia,lymphadenopathy,white blood cells(WBC),neutrophils There were no significant differences in cell percentage,erythrocyte sedimentation rate,C reactive protein(CRP)and B natriuretic peptide(BNP)between genotypes GG and GA + AA(P <0.05).Conclusions 1.There are three genotypes of GG,GA and AA at the rs2333227 locus of MPO gene,and there may be no significant correlation with the susceptibility to Kawasaki disease in Han children in Inner Mongolia.2.The MPO gene rs2333227 locus SNP may be related to Kawasaki disease complicated with CAL in Han children in Inner Mongolia.3.MPO gene rs2333227 locus GG genotype may increase the risk of Kawasaki disease and CAL in Han children in Inner Mongolia.4.MPO gene rs2333227 loci and clinical characteristics of Kawasaki disease in Han children in Inner Mongolia(rash,lip and oral manifestations,hand and foot symptoms,conjunctival hyperemia,lymphadenopathy,WBC,neutrophil percentage,There was no significant correlation between ESR,CRP and BNP,and the difference was not statistically significant(P <0.05).
