Study On The Effects Of Fuyanshu Capsules On Bax,Bcl-xL,Caspase-3,and Pbef Of PID Model Rats Based On Apoptosis Mechanism

Objective : Selecting Fuyanshu Capsules,which have achieved clinically significant effects,to observe its effects of Bax,Bcl-x L,Caspase-3 and PBEF,and PBEF m RNA expression in uterine tissues on acute pelvic inflammation model rats.To explore the mechanism of Fuyanshu Capsules in treating PID,and provide scientific basis for the clinical application of Fuyanshu Capsules in the treatment of pelvic inflammatory diseases.Methods:Forty two 8-week-old SPF female rats were selected.Six healthy rats were randomly selected for blank one week after adaptive feeding,and the remaining 36 rats were used for modeling.The method of mixed bacterial solution injection + mechanical damage was used to make the model.After 7 days of adaptive feeding,the model rats were randomly divided into 6 groups(Fuyanshu capsule high,medium and low dose group,Kangfuyan capsule group,cefixi capsule group,model group).Each group consisted of 6 rats.After 14 days of continuous treatment,the uterus of the rats was sectioned for pathology.After HE staining,the changes of the morphology of the uterus were observed under the microscope,and the degree of inflammation was graded and scored,and examined by RT-PCR the expression of PBEF m RNA in uterus was measured,and the content of Bax,Bcl-xl,Caspase-3 and PBEF in uterus was measured by Elisa.Results:(1)The score of rats’ uterine inflammatory pathological changes: Compared with the blank group,the pathological score of uterine inflammation in the model group was significantly higher(P<0.05);compared with the model group,the scores of cefixime capsule group,Kangfuyan capsule group and Fuyanshu capsule high dose group were significantly lower(P<0.05);the scores of middle dose group and low dose group of Fuyanshu capsule were lower,But the difference was not statistically significant(P>0.05).(2)The expression of Bax in uterus of rats: Compared with the blank group,the concentration of Bax in the uterus of the model group was significantly higher(P<0.01);compared with the model group,the concentration of Bax in the uterus of the rats in the model group was decreased,but the difference was not statistically significant.(3)The expression of Bcl-xl in uterus of rats: Compared with the blank group,the concentration of Bcl-xl in the uterus of the model group increased significantly(P<0.05);compared with the model group,the concentration of Bcl-xl in the uterus of the high-dose Fuyanshu capsule group decreased significantly(P There was no significant difference between the other groups(P>0.05).(4)The expression of Caspase-3 in uterus of rats: Compared with the blank group,the concentration of casp-3 in the uterus of the model group was lower,the difference was statistically significant(P<0.05);compared with the model group,the concentration of Caspase-3 in the uterus of the high dose Fuyanshu capsule group and Kangfuyan capsule group was higher,the difference was statistically significant(P<0.05)The concentration of Caspase-3 in the uterus of the rats in the other groups showed an increasing trend,with no significant difference(P>0.05).(5)The expression of PBEF in uterus of rats: Compared with the blank group,the concentration of PBEF in the uterus of the model group increased significantly(P<0.05);compared with the model group,the concentration of PBEF in the uterus of the high dose Fuyanshu capsule group,Kangfuyan capsule group and cefixi capsule group decreased significantly(P<0.05),the difference was statistically significant(P<0.05),and the concentration of PBEF in the uterus of the other groups decreased There was no significant difference(P>0.05).(6)Expression of PBEF m RNA in uterus of rats: Compared with blank group,PBEF m RNA expression in uterus of model group was significantly higher(P <0.01);compared with model group,PBEF m RNA expression in uterus of cefixi capsule group,Kangfuyan capsule group and high dose Fuyanshu capsule group was significantly lower(P<0.05);PBEF m RNA expression in uterus of low dose and medium dose Fuyanshu capsule group was significantly lower(P<0.05)There was no significant difference in PBEF m RNA expression.(7)Correlation analysis: selectting the experimental data of each group for correlation analysis,as shown in Table 12,the expression of Bax protein is positively correlated with the expression of Bcl-xl protein,the correlation coefficient = 0.381,the correlation is statistically significant(P<0.05);the expression of PBEF is negatively correlated with the expression of Caspase-3,the correlation coefficient =-0.364,the correlation is statistically significant(P<0.05);there was no significant correlation between the other indexes(P>0.05).Conclusions:1.In this experiment,the method of mixed bacterial solution injection + mechanical injury was used to establish the model.The pathological score of uterine inflammation in the model group was significantly increased,and a large number of neutrophils infiltration was seen under the microscope of the uterine tissue section,indicating that the model was successful;2.Fuyanshu capsule can reduce the score of pathological changes of uterine tissue inflammation in PID model rats and reduce the inflammation of uterus in rats,which shows that Fuyanshu capsule has therapeutic effect on PID;3.The pathogenesis of pelvic inflammatory disease may be related to apoptosis.Fuyanshu capsule can alleviate the inflammatory pathological changes of rats’ uterus.The mechanism may be through reducing the expression of PBEF,at the same time,down regulating the expression level of Bcl-xl protein and up regulating the expression of Caspase-3,so as to regulate the apoptosis of inflammatory cells and finally achieve the effect of treating PID.
