Objective:To investigate the mechanism of Qianjintablet to treat rats with pelvic inflammatory disease.Methods:Wistar female rats were selected to establish acute pelvic inflammatory disease model by vaccinating mixed bacterial liquid in uterus.120 experimental rats were randomly divided into the blank group, the sham-operation group, the model group, the low dosage group of Qianjintablet, the middle dosage group, the high dosage group, the Fuyankang group, the Roxithromycin group. After 21 days of drug treatment,rats'tissues such as uterus,uterine tube,ovary were collected. Analysing peripheral blood cells by biochemistry analyzer; Testing the content of IgA,IgG,IgM,IL-2,IL-6,IL-8,TNF-α,NF-kB by ELISA; Observing pathological change of rats'uterine tube,ovary by electric mirror; Testing the expression of ICAM-1,Caspase-3,MMP-2,Bax,Bcl-2 in rats'uterine tube,ovary by immunohistochemisty.Results:1. blood cells:the number of the WBC, Neutrophil, Lymphocytes, Monocyte in every group were higher than the model group (P<0.05).2. IgA,IgM,IgG:the content of IgA,IgM, IgG in the lower,middle, high dosage group were separately higher than the model group (p< 0.05); the high dosage group, the Fuyankang group, the Roxithromycin group were separately higher than the lower,middle dosage group (p <0.05)3. the content of cytokine:IL-2:the lower,middle,high dosage group was higher than the model group(p<0.05); IgG,IgM,IL-2,IL-6,IL-8,TNF-α,NF-kB:every treating group was obviously higher than the model group (p<0.05)4.ICAM-1,Caspase-3,MMP-2,Bax,Bcl-2:(1) ICAM-1,caspase-3:The treating groups'positive cells number of mouse's tissues like uterus,uterine tube,ovary were lower than the model group (P< 0.05); the high dosage group of Qianj intablet, Fuyankang group, the Roxithromycin group's positive cells number of mouse's tissues like uterus,uterine tube,ovary were lower than the low dosage group of Qianj intablet and the middle dosage group of Qianjintablet (P<0.05)(2)MMP-2:The high dosage group of Qianjintablet, Fuyankang group, the Roxithromycin group's MMP-2 gray threshold of mouse's tissues like uterus,ovary was higher than the model group(P<0.05); the high dosage group of Qianjintablet, Fuyankang group, the Roxithromycin group's MMP-2 gray threshold of mouse's tissues like uterus,uterine tube,ovary was lower than the low dosage group of Qianjintablet and the middle dosage group of Qianjintablet (P<0.05) (3)Uterus Bax,Bcl-2:Bax:the low dosage group of Qianjintablet, the middle dosage group of Qianjintablet, Fuyankang group's Bax positive cells number were significantly lower than the model group (P<0.05); Bcl-2:the treating group was significantly higher than the model group (P<0.05)5.The results of SEMThe intimal epithelium cells of model group which in mouse's tissues like uterus,uterine tube,ovary had falled off, degenerated, fibroblast proliferated. The intimal interstitial cells of mouse's terus,ovary and oviduct in the low dosage group of Qianjintablet, the middle dosage group of Qianjintablet, the high dosage group of Qianjintablet was normal expect mitochondria edema and vacuolated.Conclusion:Qianjintablet may play an anti-inflammatory in adjusting the peripheral white blood cells,promoting the formation of immunoglobulin, adjusting the expression and content of inflammation factor in pelvic tissue, Qianjintablet may be effective in treating pelvic inflammatory disease.
