| objective: Making chronic pelvic inflammatory disease models with rats by phenol mucilage method,to study one part of the treatment mechanism of Jia Lian pelvic Capsule(produced in our hospital)what used to cure pelvic inflammatory disease,and to provide a better method for the clinical application of the medicament on the molecular level.Methods : Selected 80 female SD rats,divided them into normal group,sham operation group,model group,Fukeqianjin capsule group and JiaLian pelvic capsule group.normal group without any treatment;the sham operation group with sterile saline instead of phenol mucilag;model group,Fukeqianjin capsule group,Jia Lian pelvic Capsule group were used to make the model of chronic pelvic inflammatory by phenol mucilage method.After the success of the building,drug treatment for 14 days by gavage.Then aortic blood collected from the rat's abdominal aorta and took the left side uterus,Fallopian tubes of the rats.The uterus,Fallopian tubes tissue for the preparation of pathological section and HE staining,and then observed the changes of morphological;blood samples were centrifuged to extract serum samples,then detected IL-2,TNF-? of each experimental groups by ELISA.Results: the normal group and the sham group compared with the model group,the difference of the serum inflammatory factors was statistically significant(p <0.01),and the normal group compared with the sham group in serum inflammatory factors(p> 0.05),the difference was not statistically significant,showing that the rat models with chronic pelvic inflammatory building success.1)Pathological Results: form the macroscopic observation,model group :the pelvic cavity was congestion,uterus and Fallopian tubes tissue were swelling and congestion,the form of organization was changed,and some tissues were obvious adhesion with pelvic tissues;FuKeqianjin capsule group and the JiaLian pelvic capsule group: the pelvic cavity was basic normal,there was no obvious congestive,the texture of uterus and Fallopian tubes tissue was normal,individual tissue had congestion,edema,and pelvic cavity had no obvious adhesion.Form the HE staining observation: the model group: uterine wall structure layer was disorder,endometrial congestion and edema,epithelial cell was proliferation,degeneration,necrosis and shedding,inherent layer had a lot of inflammatory cells,some glandular cells were necrotic,uterine smooth muscle fiber was disordered.FuKeqianjin capsule group and the JiaLian pelvic capsule group: uterine wall tissue structure was basic clear,endometrial epithelial cells arranged basic neat,showing a few cells of degeneration,necrosis and shedding,the glands were completed,there were a few inflammatory cells,smooth muscle fibers were smooth,there was no obvious adhesion,percreta was integrated.2)IL-2: FuKeqianjin capsule group compared with the model group decreased,the difference was statistically significant(p <0.05);the JiaLian pelvic capsule group compared with the model group decreased,the difference was statistically significant(p <0.01).3)TNF-? : FuKeqianjin capsule group compared with the model group decreased,the difference was statistically significant(p <0.01);the JiaLian pelvic capsule group compared with the model group decreased,the difference was statistically significant(p <0.01).Conclusion: The rat model with chronic pelvic inflammatory was builded successful by phenol mucilage;The mechanism of the treatment of chronic pelvic inflammatory disease using JiaLian pelvic Capsulein regulating or FuKeqianjin capsule in serum IL-2 and TNF-? may be similar;the JiaLian pelvic Capsule in curing chronic pelvic inflammatory rat model may play a role through regulating rats' serum inflammatory factors and repairing the form of organization of uterus and Fallopian of rats.JiaLian pelvic Capsule may can regulate the level of serum inflammatory factors,and then regulate the immune mechanism to control the progress of inflammation,relieve disease symptoms. |
