Objective:To detect the dynamic changes of dendritic cells and cytokines HMGB-1,NKG2 D,IFN-? and TNF-? in peripheral blood of patients with non-small cell lung cancer during chemotherapy;Analysis the correlation between dendritic cells and HMGB-1,NKG2 D,IFN-? and TNF-?.Method:A total of 45 patients who were initial diagnosed as inoperable non-small cell lung cancer in the Oncology Department of our hospital from September2018 to October 2019 were selected,all of whom successfully completed two chemotherapy cycles.The fasting peripheral venous blood was collected from the included patients before the first cycle of chemotherapy,on the 20 th day of the first cycle of chemotherapy,and on the 20 th day of the second cycle of chemotherapy.The level of HMGB-1,NKG2 D,IFN-? and TNF-? were determined by enzyme-linked immunosorbent assay(ELISA).The count of DC cells,the number of living DC cells and the survival rate of DC cells were measured by immunomagnetic beads.The indicators above were analyzed by Spss21.0 software.The changes of the indicators before and after chemotherapy were compared by repeated measurement analysis of variance,respectively.The correlation between DC and HMGB-1,NKG2 D,IFN-?,TNF-? before and after chemotherapy were analyzed by pearson correlation analysis,respectively.Result:1.Changes in the concentration of HMGB-1,NKG2 D,IFN-? and TNF-?in peripheral blood during chemotherapyThe concentration of HMGB-1,NKG2 D,IFN-? and TNF-? decreased gradually during the period of chemotherapy.The F value were 318.862 ?89.435 ? 136.537 ? 527.724,respectively,the differences were statistically significant(P<0.05).2.Changes of DC cell count,number of living DC cells and survival rate of DC cells in peripheral blood before and after chemotherapyThere was no significant difference in DC cell count before and after chemotherapy(P > 0.05),the F value was 0.237.There was no significant difference in the number of living DC cells before and after chemotherapy(P> 0.05),the F value was 0.175.There was no significant difference in the survival rate of DC cells before and after chemotherapy(P>0.05),the F value was 0.570.3.Correlation between DC and HMGB-1,NKG2 D,IFN-? and TNF-?.There was no correlation between DC and HMGB-1,NKG2 D,IFN-?,TNF-?before the first cycle of chemotherapy(P > 0.05),the Pearson correlation coefficient were-0.056,-0.084,-0.211 and 0.056,respectively.There was no correlation between DC and HMGB-1,NKG2 D,IFN-?,TNF-? after the first cycle of chemotherapy(P > 0.05),the Pearson correlation coefficient were-0.017,-0.125,0.090 and-0.046,respectively.There was no correlation between DC and HMGB-1,NKG2 D,IFN-?,TNF-? after the second cycle of chemotherapy(P > 0.05),the Pearson correlation coefficient were 0.053,-0.035,-0.120 and-0.156,respectively.Conclusion:1.In peripheral blood of patients with non-small cell lung cancer,the concentrations of HMGB-1,NKG2 D,IFN-? and TNF-? were trended downward after chemotherapy.2.Chemotherapy had no significant effect on the DC cell count,the number of living DC cells and the survival rate of DC cells in peripheral blood of patients with non-small cell lung cancer.3.There was no correlation between DC and cytokines(HMGB-1,NKG2 D,IFN-? and TNF-?)in peripheral blood before and after chemotherapy.
