| Objective NKG2D is an important activating receptor expressed by natural kill cells(NK cells),which play a significant roles on NK cells in activation, immunosurveillance and Cytotoxicity.Present research show that NK cells in chronic myeloid leukemia(CML) Compared with in normal human exist significant differences on the function,The levels of NKG2D expression were significantly lower than those in normal.Interferon-α(IFN-α),a traditional medicines to treat CML,which has been widely used for immunoregulatory in CML since 1980.The cytokine interleukin-15(IL-15) is the most important physiological growth factor,It has a key role in promoting survival,proliferation and activation of NK cells.In this study,IFN-αand IL-15 were used to improve NKG2D expression and activate NK cell in CML patients.Methods Peripheral blood mononuclear cells(PBMC) were isolated from normal adult and Newly diagnosed chronic myelogenous leukemia patients(not complete remission) by Ficoll-Hypaque density gradient centrifugation.NK cells were separated from PBMC by MACS and NKG2D expression were detected by flow cytometer.Using IFN-α,IL-15,IFN-αcombind with IL-15 cocultured with NK cells 36 hours respectively,then,NKG2D expression were detected by flow cytometer again,supernatants were harvested for IFN-γconcentration measured by ELISA kits.Cytotoxicity of NK cells was assayed by MTT method.Results The levels of NKG2D expression,secretion of IFN-γand Cytotoxicity of NK cells in Newly diagnosed CML chronic phase patients were significantly lower than those in normal controls,but after stimulated by IFN-αcombind with IL-15,the levels of NKG2D expression,secretion of IFN-γand Cytotoxicity of NK cells were striking increased which were to get close normal controls(P＜0.05)Conclusions Both IFN-αand IL-15 can not only improve NKG2D expression on NK cell in CML patients but also activate NK cell and enhance its Cytotoxicity effectively.Taken in conjunction with a stronger effect.
