Objective:Using the ELISA to detect the dynamic changes of NK cell receptors NKG2 D and IL-12,IL-15 and IL-18 in peripheral blood of patients with non-small cell lung cancer before and after chemotherapy.Analysis of the correlation between NKG2 D and IL-12,IL-15,IL-18;Explore the effect of chemotherapy on immune function in patients with non-small cell lung cancer and its clinical significance.Method:Patients with the initial diagnosis of lung cancer who were admitted to the Oncology Department of the Affiliated Hospital of Chengde Medical College from September 2018 to January 2019 were selected.The screening was performed according to the inclusion criteria and exclusion criteria,and 30 patients with non-small cell lung cancer confirmed to be inoperable by histopathology and imaging were included.Patients who failed to complete 2 cycles of chemotherapy and did not retreat after 2 cycles of chemotherapy were excluded,and 26 patients who successfully completed 3 cycles of chemotherapy were selected for analysis.Chemotherapy regimens include pemetrexed + cisplatin / carboplatin,gemcitabine + cisplatin / carboplatin,paclitaxel + cisplatin / carboplatin.Among the 26 patients who were finally included in the results analysis,14 were male and 12 were female.The pathological types were 16 adenocarcinoma,8 squamous cell carcinoma,and 2 adenosquamous carcinoma.Peripheral venous blood before the first cycle of chemotherapy,before the second cycle of chemotherapy,and before the third cycle of chemotherapy was enrolled in the enrolled patients.The concentration of NKG2 D,IL-12,IL-15,and IL-18 was detected by the ELISA method.Statistical analysis was performed using spss21.0 software.The measurement data were expressed as mean ± standard deviation.The analysis method used repeated analysis of variance analysis to compare the changes in the measured indicators before and after different cycles of chemotherapy.The correlation between NKG2 D and IL-12,IL-15,IL-18 was analyzed by person correlation.With?=0.05 as the test standard,the difference of P<0.05 was statistically significant.Result:1.Changes in NKG2 D,IL-12,IL-15 and IL-18 levels after chemotherapy The concentrations of NKG2 D before and after chemotherapy were 199.95±31.03,159.24±10.49,and 135.82±2.77,respectively.The F value was 71.775,P<0.001,and the difference was statistically significant.The concentrations of IL-12 before and after chemotherapy were 10.13±1.14,8.91±1.05,6.73±1.11,and the F value was 57.396,P<0.001.The difference was statistically significant.The concentration levels before and after IL-15 chemotherapy were 4.62±0.95,3.19±1.19,2.42±0.65,and the F value was 39.172,P<0.001.The difference was statistically significant.The concentrations of IL-18 before and after chemotherapy were 134.18 ±6.62,92.55±6.64,55.78±6.77 and the F value was 917.324,P<0.001.The difference was statistically significant.The levels of NKG2 D,IL-12,IL-15 and IL-18 were significantly lower after chemotherapy than before chemotherapy and continued to decrease with the progress of chemotherapy. 2.Correlation between NKG2 D and IL-12,IL-15,IL-18The Pearson phase coefficients between NKG2 D and IL-12,IL-15 and IL-18 before the first cycle of chemotherapy were 0.078,0.03,and-0.038,respectively;the P values were 0.706,0.886,and 0.852,respectively.The Pearson phase coefficients between NKG2 D and IL-12,IL-15,and IL-18 before the second cycle of chemotherapy were 0.062,-0.390,and 0.397,respectively;the P values were 0.765,0.049,and 0.045,respectively.The Pearson phase coefficients between NKG2 D and IL-12,IL-15 and IL-18 before the third cycle of chemotherapy were-0.091,0.473,and 0.215,respectively;the P values were 0.658,0.015,and 0.291,respectively.This indicates that NKG2 D has a correlation with IL-15 and IL-18 in the second cycle(P<0.05);NKG2D was associated with IL-15 in the third cycle(P<0.05).Conclusion:1.After chemotherapy,the levels of NKG2 D,IL-12,IL-15 and IL-18 in peripheral blood of patients with non-small cell lung cancer were reliefed,and the immune function of patients with non-small cell lung cancer was reduced after chemotherapy.2.NKG2 D has an absolute correlation with IL-15 and IL-18 after different cycles of chemotherapy;NKG2D does not correlation with IL-12.
