Aim:This study was retrospective analysis of patients with lung cancer in peripheral blood DC cells and related cytokines(IFN-r,TNF-a,NKG2 D,HMGB-1)in the dynamic change before and after radiotherapy,the overall evaluation of effect of radiotherapy on the immune function and mechanism analysis of DC cells and related cytokines(IFN-r,TNF-a,NKG2 D,HMGB-1)the relationship between the specific regulatory effect of radiotherapy on the immune function of patients with lung cancer,the best time of radiotherapy combined with immune therapy to explore the breakthrough point,provided theoretical basis for the clinical reliability for longer overall survival of patients with lung cancer.Method:To learn or cytological diagnosis of oncology Affiliated Hospital of Chengde Medical College from June 2017 to January 2018 menstrual pathological lung cancer screening of patients,30 cases of lung cancer patients were selected according to the criteria,all patients were treated with Elekta linac,using intensity-modulated radiation therapy(IMRT),Dt 60 Gy,spiral CT,2Gy\ times,5 times a week.Collect the patients before radiotherapy,radiotherapy 1 weeks,4 weeks and 2 weeks of radiotherapy radiotherapy 6 weeks fasting venous blood in-80 ?refrigerator,ELISA,IFN-r,TNF-a,NKG2 D detection and HMGB-1 detection,DC cell count,immunomagnetic beads live DC cell number,DC number of apoptotic cells and DC cell activity.Using SPSS19.0 statistical software,the measurement data were expressed by mean standard deviation.The variance analysis of repeated measurement design was used to analyze the changes of the indexes measured at different time points of patients at different time points,and person correlation analysis was also conducted.Using a=0.05 as the test standard,There is significant difference of P is less than or equal to 0.05.Result:1.4 time periods before and after radiotherapy(1 weeks before radiotherapy,2 weeks for radiotherapy,4 weeks for radiotherapy and 6 weeks for radiotherapy)showed significant differences in IFN-r,TNF-a,HMGB1,NKG2 D and DC cell counts,DC cell count and DC apoptosis cell number(P < 0.05).2.The number of IFN-r and DC cells increased gradually.TNF-a,HMGB1 and NKG2 D showed a decreasing trend.The number of live DC cells increased first and then decreased.The highest number of cells was observed at 4 weeks after radiotherapy,and the number of DC apoptotic cells showed no obvious change.The highest was at 4 weeks.3.There was no significant difference in the activity of DC cells.Conclusion:1.conventional radiotherapy promotes the expression of immuno factor INF-r and inhibits the expression of TNF-a,HMGB-1 and NKG2 D,and then activates the immune function.2.conventional radiotherapy promotes the maturation of DC cells,and the effect is most obvious at 4 weeks,promoting the action of tumor antigen.3.conventional radiotherapy has no effect on the activity of DC cells.4.There was no correlation between DC cells and related immune cell factors(INF-r,TNF-a,HMGB-1,NKG2D).
