| Objective: Observe the effect of "Tiao Shen Chang Qing" acupuncture on the expression of TLR4 and CXCR4 in the brain and colon tissues of rats with gastrointestinal dysfunction in Parkinson's disease,and explore the possibility of "Tiao Shen Chang Qing" acupuncture intervention on Parkinson's gastrointestinal dysfunction Mechanism.Method: Select 80 male SPF grade SD rats,weighing 200±20g,and divide them into blank group,sham operation group and 56 model groups according to the random number table method.For the model group,western medicine group,conventional acupuncture group and "Tiao Shen Chang Qing" group,10 rats in each group.In the modeling stage,the rotenone solution with a drug concentration of2 mg/kg was injected intraperitoneally at a dose of 2.5 ml/kg to establish a model of gastrointestinal dysfunction in Parkinson's disease;the sham operation group used the same dose of 1.25% chloroform and 4% sodium carboxymethyl cellulose solution was injected intraperitoneally.In the treatment stage,the western medicine group: give 75 mg/kg of metoprolol in combination with 2 mg/kg of domperidone,and then perform intragastric intervention treatment at a dose of 5 ml/kg.The sham operation group: give 5 ml/kg physiological saline for intragastric intervention treatment.Conventional acupuncture group: Select the "dancing tremor control zone" for Parkinson's disease according to the "Acupuncture Point Map of Experimental Animals",plus the three acupuncture points "Zusanli"according to the 13 th Five-Year Textbook of "Acupuncture" ","Zhongwan","Tianshu" for acupuncture;"Tiaoshen Changqing" group: select some acupuncture points of Professor Wang Shun's "Tiaoshen Changqing"acupuncture method,Baihui will penetrate the sun;Zhongwan,Qihai;Shenmen,Taichong,Sanyinjiao acupuncture;no intervention in the model group and the blank group.Observation indicators: After the model is built,observe the behavior scores of rats in each group,the moisture content of stool,and the residual rate of gastric contents;after two weeks of treatment,observe the behavior scores of rats,moisture content of stool,and gastric contents Residual rate and immunohistochemistry were used to detect the content of TLR4 and CXCR4 in rat brain and colon tissue;Western Blot was used to detect the content of TLR4 in colon tissue.The data statistical method uses Microsoft excel 2016 for database processing,and then uses SPSS25.0 for statistical processing.Results:1.Compared with the blank group,the behavioral score,fecal moisture content,and gastric content residual rate of the model rats were statistically significant after four weeks(P<0.05),and after 2 weeks of treatment,Compared with the model group,the "Tiao Shen Chang Qing" group,Western medicine group and conventional acupuncture group had statistically significant results in terms of academic score,fecal moisture content and gastric content residual rate(P<0.05);and Compared with the conventional acupuncture group,the results of academic score,water content of feces and residual rate of gastric contents in the "Tiao Shen Chang Qing" group and the Western medicine group were all statistically significant(P<0.05).Compared with the western medicine group,the results of the "Tiao Shen Chang Qing" group were not statistically significant in terms of academic scores,fecal moisture content,and gastric content residual rate(P>0.05).2.The content of TLR4 in the brain and colon tissues of rats in each group after 2 weeks of treatment: Compared with the blank group,there was no significant change in the content of TLR4 in the brain and colon tissues of the sham operation group(P> 0.05).The content of TLR4 in colon tissue was significantly increased(P <0.05);compared with the model group,the content of TLR4 in the brain and colon tissue of rats in the western medicine group,"Tiao Shen Chang Qing" group,and conventional acupuncture group were significantly reduced(P<0.05),and the TLR4 content of the Western medicine group was significantly lower than that of the "Tiaoshen Changqing" group and the conventional acupuncture group(P<0.05);The content of TLR4 in rat brain and colon tissue was significantly reduced(P<0.05)3.The content of CXCR4 in the brain and colon tissues of rats in each group after 2 weeks of treatment: Compared with the blank group,there was no significant change in the content of CXCR4 in the brain and colon tissues of the sham operation group(P>0.05),and the content of CXCR4 in the model group was in the brain Significantly increased in the tissue(P<0.05),and the content in the colon tissue was significantly reduced(P<0.05);compared with the model group,"the conventional acupuncture group CXCR4 content in brain and colon tissue was not significantly different(P>0.05).The content of CXCR4 in the Tiaoshen Changqing group and the western medicine group were significantly reduced in the brain tissue(P <0.05),and the content in the colon tissue were significantly increased(P<0.05).There was no significant difference in the content of CXCR4 in the brain and colon tissue between the Qing group and the Western medicine group(P>0.05);compared with the conventional acupuncture group,the content of CXCR4 in the Tiao Shen Chang Qing group and the Western medicine group were significantly reduced(P<0.05),the contents in the colon tissue were significantly increased(P<0.05)Conclusion:(1)PD gastrointestinal dysfunction model rats showed that the content of TLR4 in brain and colon tissues increased significantly;the content of CXCR4 in brain tissue increased significantly,and the content of CXCR4 in colon tissue decreased significantly.(2)"Tiaoshen Changqing" acupuncture treatment can significantly inhibit the expression of TLR4 in the brain and colon of rats,and the effect is better than the conventional acupuncture group,which is basically the same as the Western medicine group;"Tiaoshen Changqing" acupuncture method can inhibit CXCR4 in the brain The expression in tissues and the promotion of CXCR4 expression in the colon are comparable to Western medicine treatment,and the effect is better than conventional acupuncture treatment.(3)In terms of improving the behavioral score of PD with gastrointestinal dysfunction,the water content of feces in rats and the residual rate of gastric contents,"Tiao Shen Chang Qing" acupuncture treatment can almost achieve the effect of Western medicine treatment,and is superior to conventional acupuncture
