Effects Of "Tiaoshen Changqing" Acupuncture On ?-syn In Different Parts Of PD With Gastrointestinal Dysfunction Model Rats

Objective:The purpose of this experimental study was to clarify Parkinson's disease by observing the changes in motor function,fecal water content,gastric content residual rate,the content of ?-syn in midbrain substantia nigra,gastric and colon tissue in rotenone-induced PD with gastrointestinal dysfunction model rats,to reveal the effect of "Tiaoshen Changqing" acupuncture on PD with gastrointestinal dysfunction symptoms and explore its related pathogenesis.Methods:SPF adult SD rats were randomly divided into six groups according to the random number table method:including blank group,sham operation group,model group,western medicine group,Tiaoshen Changqing group and conventional acupuncture group.PD with gastrointestinal dysfunction model rats were established by intraperitoneal injection of rotenone solution for 5d/week for 4 weeks;model detection was tested by the APO-induced climbing experiment.Experimental intervention methods: According to the "Tiaoshen Changqing" acupuncture method proposed by Professor Wang Shun,the Tiaoshen Changqing group selected the "Baihui" and " Temple" of the head acupoint,the "Zhongwan" and "Qihai" of the abdominal acupoint,The "Shenmen","Sanyinjiao" and "Taichong" acupoints of the three Yin Channels are intervened with acupuncture.The conventional acupuncture group selected "Dance tremor control zone" and "Tianshu","Zusanli" and "Zhongwan" acupoints for intervention treatment.Once a day,continuous treatment for 2 weeks.The western medicine group was given Metopar combined with Domperidone for intragastric intervention treatment,once a day for 2 weeks.The blank group,sham operation group,and model group were fed normally without treatment intervention.Observe the motor function and weight change of rats in each group,test the water content of feces,the residual rate of gastric contents,immunohistochemistry and enzyme-linked immunosorbent assay were used to detect the expression of ?-syn in the substantia nigra,stomach and colon tissues of rats.Realuts:1.In terms of weight: there was no significant statistical difference in weight between the blank and sham operation group of rotenone-induced PD with gastrointestinal dysfunction model rats(P>0.05);After treatment,compared with the model group,the weight of rats in the western medicine group and the two-acupuncture group were all increased,In addition,the weight gain of rats in Western medicine group and Tiaoshen Changqing group was larger(P<0.01),Routine acupuncture group followed(P<0.05).2.In terms of autonomous motor function: before intervention treatment,the autonomous activity of rats in each model group was significantly lower than that in the blank group(P <0.01);after intervention treatment,the movement symptoms of rats in the western medicine group and the two-acupuncture group were obvious improved,which was statistically significant compared with the model group(P <0.01);the improvement of the motor symptoms of the western medicine group was better than the Tiaoshen Changqing group(P <0.05),and the Tiaoshen Changqing group was better than the conventional acupuncture group(P <0.05).3.In terms of the water content of feces: before intervention treatment,the water content of feces of the model rats was significantly lower than that of the blank group,with a statistical difference(P<0.01);After intervention treatment,the water content of feces in the western medicine group and Tiaoshen Changqing group increased,followed by the conventional acupuncture group,but there was a statistical difference compared with the model group(P<0.05);4.In terms of gastric content residual rate: before interventional treatment,the gastric content residual rate of model rats increased significantly compared with the blank group,with statistical difference(P<0.05);After intervention treatment,the residual rate of gastric contents in the western medicine group and Tiaoshen Changqing group decreased,followed by the conventional acupuncture group,but there were statistical differences compared with the model group(P <0.05);5.In terms of small bowel advancement rate: The small bowel advancement rate of rats in model group was significantly lower than that of the blank group(P<0.01);after intervention treatment,the rate of small bowel advancement in the western medicine group and the two-acupuncture groups was significant improved,which was sta-Resluts: tistically different from that of the model group(P<0.01);in addition,the improvement in the western medicine group was superior to the Tiaoshen Changqing group,and the conventional acupuncture group was inferior to the Tiaoshen Changqing group;6.Changes of ?-syn in substantia nigra: In the blank group and the sham-operated group,there was no obvious abnormal cell morphological structure and abnormal aggregation of ?-syn.In the model group,?-syn positive cells increased,and the concentration of ?-syn was higher than that in the blank group(P<0.01).After intervention treatment,the Western medicine and the two-acupuncture groups can reduce the concentration of ?-syn,which was statistically significant compared with the model group(P<0.01).The effect of Western medicine group is better than Tiaoshen Changqing group(P<0.05),the effect of Tiaoshen Changqing group is better than conventional acupuncture group(P<0.05).7.Changes of ?-syn in stomach and colon tissue: In the blank group and the sham-operated group,there was no obvious abnormal cell morphological structure and abnormal deposition of ?-syn.However,there was an increase in ?-syn positive cells in the gastrointestinal tissues of rats in each model group,and the concentration of ?-syn was significantly higher than that in the blank group,with a significant statistical difference(P<0.01).After intervention treatment,the concentration of ?-syn in gastric and colon tissues of rats in western medicine group and two-acupuncture group was statistically different from that in model group(P<0.01);in addition,the the improvement of Western medicine group was superior to the Tiaoshen Changqing group,and the conventional acupuncture group was inferior to the Tiaoshen Changqing group.Conclusion:1.In the PD with gastrointestinal dysfunction model rats,the substantia nigra,stomach,and colon tissues all showed abnormal expression and aggregation of ?-syn,and also showed autonomous motor dysfunction,decreased fecal water content,increased gastric content residual rate and gastrointestinal emptying significantly slowed performance.2."Tiaoshen Changqing" acupuncture treatment can significantly inhibit the ab-normal expression and aggregation of ?-syn in the substantia nigra,stomach and colon tissues of PD with gastrointestinal dysfunction model rats,and the effect of "Tiaoshen Changqing" acupuncture treatment is better than conventional acupuncture,which,however,is similar to Western medicine.3."Tiaoshen Changqing" acupuncture treatment can significantly improve the autonomic motor function,increase the water content of feces,reduce the residual rate of gastric contents and accelerate the advancement rate of small intestine of PD with gastrointestinal dysfunction model rats,and the effect of "Tiaoshen Changqing" acupuncture intervention is better than conventional acupuncture methods,which is similar to Western medicine.4.One of the mechanisms of "Tiaoshen Changqing" acupuncture treatment of PD gastrointestinal dysfunction may be achieved by reducing the abnormal expression of ?-syn in the substantia nigra,stomach and intestine tissues.
