Androgenic alopecia(AGA),also known as male alopecia or seborrheic alopecia,is one of the common types of alopecia.AGA is an autosomal dominant genetic disease,and androgen dependent microminiaturization of hair follicles caused by increased concentration of dihydrotestosterone in the scalp is one of the important pathogenic factors.Although there are many studies on AGA,only finasteride and minoxidil are recommended by the United States food and Drug Administration(FDA).Finasteride is a 5 ?-reductase inhibitor,it can inhibit the transformation of testosterone(T)to dihydrotestosterone(DHT),which has a stronger affinity for androgen receptors,and provides an effective target for the treatment of AGA.However,two-thirds of the patients have different degrees of sexual dysfunction in the course of using finasteride,which is often manifested as hyposexual desire,hyposexual function,decreased ejaculation,and ejaculation disorder.Minoxidil is recognized as a topical therapeutic drug of AGA,and its mechanism of action is not clear.In the process of clinical application,adverse reactions such as contact dermatitis,skin irritation,dizziness and tachycardia were also observed,and hair loss may continue to progress after discontinuation.In conclusion,the efficacy and safety of finasteride and minoxidil are not very satisfactory.Therefore,it is of great social and clinical significance to study new effective treatment methods.Objective:To explore the therapeutic effect and possible mechanism of Impatiens balsamina,Lawsonia inermis L and Henna on androgenic alopecia by using C57BL/6 mouse model.Methods:?Male C57BL/6 mice were used to construct androgenic alopecia model;?The animals were randomly divided into model group(M),blank control group(K)and drug group(T).The drug group was divided into T1(Impatiens balsamina group),T2(Lawsonia inermis L group),T3(Henna group)and T4(minoxidil group)according to different Experimental drug.After depilation of the central area of the back,all mice were treated with different drugs;?After 35 days of treatment,the images of the experimental areas were collected,the concentrations of T,DHT in serum and T,DHT,5alpha-reductase type 2(5aR2)in the skin samples of the experimental areas were measured,and the whole skin samples of the experimental areas were taken for histopathological examination;?SPSS 20.0 software was used to analyze the experimental data.Results:?The male C57BL/6 mice model of androgenic alopecia was established successfully;?After 35 days of treatment,new hair was found in the experimental area of mice in Group T;?The total number of hair follicles(22.75±4.301,31.38±13.212,33.00±13.825,31.12±8.357)and the number of hair follicles(13.25±2.605,18.88±9.463,21.25±8.379,21.62±5.805)in Group T were higher than those in Group M(9.12±6.854,3.62±2.504),the difference was statistically significant(P<0.05);?The concentrations of T(0.81±0.12,0.85±0.19),DHT(1.41±0.23,1.29±0.39)in the skin of Group T1 and T3 were significantly lower than those of Group M(4.75±0.41,6.48±0.57),the difference was statistically significant(P<0.05);?There was no significant difference in serum T and 5aR2 between Group T and Group M.Conclusion:?AGA mice model was successfully prepared;?The therapeutic effects of Impatiens balsamina,Lawsonia inermis L and Henna were observed in androgenic alopecia mice model;?Impatiens balsamina and Henna may play a therapeutic role by reducing the androgen concentration in skin tissue.
