Studies On Saponins From The Seeds Of Impatiens Balsamina L.

Impatiens balsamina L., a well-known herbal medicine,is now widely cultivated as a medicinal and ornamental plant in most provinces of China. The seeds, semen impatientis, have been used to treat lump in the abdomen and cancer of the esophagus, to act as an emmenagogue, to suppress puerperal pain, etc. The seeds have been also used as expectorant, bactericide, anticancer agent. So, it is valuable to do reseach on the seeds of Impatiens balsamina L..The naphthas and fattinesses in this plant have been reported more before, but the saponins have been reported fewer in China. For the sake of the better use of this herb medicine, this paper describes the extraction, isolation, identification and the contents determination of the saponins from the dried seeds of Impatiens balsamina L.. Extraction and SeparationDried seeds (5kg) were extracted with hot water (75, 60, and 45L respectively) for three times (3, 2 and 1h respectively). The extracted solution was absorbed by macroporous adsorption resin, then eluted with 95% EtOH. The extraction was subjected to silica gel column chromatography using methanol-ethylacetate and methanol -chloroform as mobile phase repeatedly to afford compounds JA,JB,JC and JD. The extraction ( compounds JD,JB and JC) was subjected to ODS column chromatography using methanol-water as mobile phase to afford J1, J2, J3, J4, J5 and J6 respectively.Compounds J1~J6 were identified respectively by IR, MS, and 1DNMR as Hosenkoside K [hosenkol A-3-O-glucopyrano -syl(2→1)-glucopyranosyl-26-O-glucopyranosyl-28-O-gluco-pyranoside] , Hosenkoside A [hosenkol A-3-O-glucopyranosyl (2→1)-glucopyranosyl-28-O-glucopyranoside], Hosenkoside M[hosenkol A-3-O-glucopyranosyl (2→1)- xylopyranosyl-26- O-glucopyranosyl-28-O-glucopyranoside], Hosenkoside L [hosenkol A-3-O-glucopyranosyl(2→1)-xylopyranosyl-28- O- glucopyranoside], Hosenkoside B [hosenkol B-3-O-glucopy- ranosyl(2→1)-glucopyranosyl-26-O-glucopyranoside] and Hosenkoside C [hosenkol C-3-O-glucopyranosyl (2→1)-glu- copyranosyl-28-O-glucopyranoside].Determination of saponins in the seeds of Impatiens balsamina L. by HPLCIn this paper we present a more sensitive quantification method by HPLC-ELSD for measuring the major saponins (compounds J1 and J2) in the seeds of Impatiens balsamina L..HPLC was performed on an Eclipse XDB-C18 column (4.6×250 mm, 5μm) using methanol and water as mobile phase (gradient elution, 0~22min,50:50; 22~50 min,54:46.) with the flow rate of 0.8 ml/min at 25℃,The gas(air) pressure of ELSD was 2.1 Bar and the temperature of drift tube for measurement was 89℃.The contents of the two saponins-compounds J1 and J2 in the seeds of Impatiens balsamina L. were determined by the above method. The results showed that the average contents of compounds J1 and J2 were 0.179 % and 0.0730 % respectively。In conclusion, All the results of this paper provided the foundations of quality standard establishment, development and utilization for Impatiens balsamina L..
