Object:With AGA mice model as the research object, the mice back external medicine composition of OA,observe of hair growth in mice and detect the serum T,E2,T/E2ratio,studying the treatment effect of OA on AGA and its related mechanism.Methods:56B6CBAF1/J male mice are numbered according to the weight.According to the random number table method to choose13mice as the blank group,the remaining mice were fed in different cages. Except the blank group,without group mice were in accordance with5mg/Kg·d dose given subcutaneous injection of testosterone propionate, continuous administration for4weeks,causing androgenic alopecia animal model.Blank group was injected with physiological saline.The fourth week of the test mice after modeling success,in the blank group and ungroup mice were randomly selected3mice were sacrificed by dislocation of the neck,to observe the pathological changes of hair loss and skin tissue.The remaining40ungrouped mice were divided into model group,medium group,positive control group,OA group,10mice in each group,continue to give the injection of testosterone propionate.At the same time,OA group with1%OA topical solution,the positive control group of5%topical minoxidil topical solution, matrix group without blank solution of OA,model group were not administered. At the end of the eighth weeks,the mice femoral artery blood serum,extraction of serum.After blood, the mice were sacrificed by dislocation of cervical, parallel to the spine,the mice back hair removal skin tissue HE staining,calculating terminal hair/vellus hair ratio.Apply Chemiluminescence micro particle immunoassay system and supporting the people of testosterone, estradiol assay kit Applied Chemistry of mouse serum testosterone,estradiol T. E2content and T7E2ratio.The measurement process a predetermined program is done automatically by the instrument.Results:1.Mice modeling two weeks later,back hair gradually began to lose their luster, modeling four weeks,eight weeks the mice appear back hair loss,gradual development from the head to the tail, and shedding intensified.The skin tissue pathology prompts hair follicle miniaturization,decline in the proportion of number of terminal hair/vellus hair. Show the AGA mice model was successfully built.2.The first eight weeks of the experiment,mice back skin HE staining,counting the number of terminal hairs and vellus hair.Ultimately terminal hair/vellus hair ratio comparison,OA group compared with model group,medium group,Increase the ratio of (P<0.01); compared with the blank group, decreased (P<0.05); No significant difference between the OA group and positive control group.3.Eight weeks of the experiment,comparison between the mice in each group of serum T,E2and T/E2proportion.Compared with the blank group, the other4mice serum testosterone, estradiol in T,E2,T/E2ratio were significantly increased (P<0.01).Comparison between groups, groups T, E2, T/E2had no significant difference (P>0.05).Conclusion:1.B6CBAF1/J mice subcutaneous injection of testosterone propionate AGA mice model is indeed valid, the effect of modeling and model stability are better.2.OA treatment of androgenetic alopecia is effective,compared with the positive control minoxidil, it promotes the treatment of hair growth.OA in the treatment of androgenetic alopecia may also reverse the proportion of terminal hairs and vellus hair, improve hair follicle miniaturization.3.OA on AGA for four weeks can not change T.E2.T/E2levels.
