Study On The Regulation Of Qi Gui Bu Xue Granule On Spleen Autophagy In Mice With Blood Deficiency

Objective: In this study,cyclophosphamide(CY)was used to induce blood deficiency in mice,to observe the regulatory effect of qiguibuxue granule on spleen autophagy of mice with blood deficiency induced by cyclophosphamide after chemotherapy,and compare the therapeutic effect of qiguibuxue granule in high,middle and low doses,so as to provide an effective basis for clinical application.Methods: After three days of adaptive feeding,formal experiments began,Cyclophosphamide was injected into the left lower abdominal cavity of mice,60 mice were randomly divided into 6 groups: blank group,model group,high,medium and low dose group of qiguibuxue granules,positive drug Siwutang group,Cyclophosphamide was injected continuously for three days,and then Chinese medicine was infused into stomach for four days.RBC,WBC,HGB and HCT were detected by hematology analyzer.Erythropoietin(EPO)was detected by ELISA.Western blot was used to detect the protein content of PI3KC3 and Beclin1.By IHC staining,Beclin 1,BCL 2 and Bax in spleen were detected.HE staining was used to detect: pathological sections of spleen.The expression level of EPO receptor gene in spleen was detected by PCR.Results:(1)The experimental results showed that qiguibuxue granule could significantly increase the hemopoietic function and influencing factors of mice with blood deficiency induced by cyclophosphamide:The content of RBC,WBC,HGB and HCT can improve the expression level of EPO in different degree;(2)The detection of related factors related to autophagy showed that qiguibuxue granule could increase PI3KC3 and decrease the expression level of Beclin 1 in the serum of blood deficiency model mice.Conclusion: The experimental results showed that(1)qiguibuxue granule had an obvious promoting effect on the hemopoietic function of cyclophosphamide induced blood deficiency model mice,and the mechanism was related to the cytokines related to the regulation of hemopoietic function of the drug;(2)Qiguibuxue granule has an obvious regulatory effect on the autophagy function of the blood deficiency model mice induced bycyclophosphamide.
