Objectives:To discus the Chinese Medicine theory liver generating blood and Qi by application of XiaoChaiHu Decoction, XiaoChaiHu and DangGui BuXue compound Decoction and ChaiHu Sheng Xue Prescription to anemia mice prepared by complex etiology methods,which will provide another treating principle and method to clinical treatment of anemia. Meanwhile,it also provide an experimental evidence for the further study of Chinese medicine theory of liver generating blood and Qi.Methods:To summarize the Chinese medicine theory liver generating blood and Qi by collating and analyzing the relating discussion searched in more than 1000 kinds of ancient Chinese medical books collected in Chinese medicine encyclopedia,the SPF male and female mice were randomly divided into blank control group, control group, XiaoChaiHu Decoction group, Danggui Buxue Decoction group, Xiaochaihu and Danggui Buxue compound Decoction group, and Chaihu Shengxue preccription group. Blood deficiency mice model were prepared by complex etiology modeling method for 14 days. At the 15th day, Decoctions were administrated to different treatment groups mice everyday, according to their advanced division,7 days for 1 course, treatment continued 2 courses. These indexes were observed?the change of body weight of mice.(2)hematopoietic cytokines content of SCF,EPO, IL-3,TGF-?1 and TNF-?:after 1 week and 2 weeks of administration,8 mice were randomly selected from each group, male and female half to half, to pick the mice eyeball for getting blood to detect blood cells and blood serum hematopoietic cytokines content of SCF, EPO, IL-3, TGF-?1and TNF-? by Elisa method.?brain, heart and liver tissue pathological changes:Later, the mice were killed to remove liver, heart and brain for making pathological sections to observe the brain, heart and liver tissue pathological changes of ischemia by evaluating tissue ischemia condition.Results:1. weight change results:After 1 week, compared with blank control group and control group, the body weight of mice had no statistically significant changes (P> 0.05). There was no statistically significant difference among treatment groups (P> 0.05). After 2 weeks, compared with the blank control group, the control group weight were less than the blank control group (P< 0.05). Compared with the control group, Danggui Buxue Decoction group, Xiaochaihu Decoction and Danggui Buxue compound Decoction group, Chaihu Shengxue prescription group mice weighted greater than control group significantly (P< 0.05). Chaihu Shengxue prescription group mice weighted greatest among other groups and the difference was significantly.(P<0.05).2. blood test results:compared with control group, blood cells were improved in different degrees in each treatment groups. After 1 week administration, RBC and HGB content in blood of Xiaochaihu and Danggui Buxue compound Decoction group, and Chaihu Shengxue prescription group were significantly higher than those in control group (P<0.05). After 2 weeks administration, RBC content in blood of Xiaochaihu Decoction and Danggui Buxue compound Decoction group was significantly higher than that of control group (P<0.05). HGB and WBC content in blood of Chaihu Shengxue prescription group was significantly higher than that of control group (P<0.05).3. detection of hematopoietic cytokines in the serum results:(1) after 1 week administration, compared with control group, EPO content in serum of Xiaochaihu Decoction group, Xiaochaihu and Danggui Buxue compound Decoction group, and Chaihu Shengxue prescription group were significantly increased (P<0.05). After 2 weeks administration, compared with the control group, EPO content in serum of Xiaochaihu and Danggui Buxue compound Decoction group, and Chaihu Shengxue prescription group were significantly increased (P< 0.05).(2) After 1 week administration, compared with control group, SCF content in serum of Xiaochaihu Decoction group were significantly increased (P<0.05). Among the other groups, there was no significant difference (P>0.05). After 2 weeks administration, difference between control group and treatment groups was not statistically significant (P>0.05).(3) Compared with control group, SCF content in serum of treatment groups was higher than that of control group, but had no significant difference (P>0.05).(4) After 1 week administration, compared with control group, TGF-?1 content in serum of treatment groups decreased, but Xiaochaihu and Danggui Buxue compound Decoction group, and Chaihu Shengxue prescription group were significantly decreased (P< 0.05). After 2 weeks administration, compared with control group, TGF-?31 content in serum of treatment groups decreased, but Chaihu Shengxue prescription group were significantly decreased (P<0.05).(5) After 1 week administration, compared with control group, TNF-a content in serum of Xiaochaihu Decoction group was significantly decreased (P<0.05). After 2 weeks administration, compared with control group, Danggui Buxue Decoction group, Xiaochaihu Decoction group, TNF-a content in serum of Danggui Buxue Decoction group, Xiaochaihu Decoction group, Xiaochaihu and Danggui Buxue compound Decoction group, and Chaihu Shengxue prescription group was significantly decreased (P<0.05).4.Brain, heart and liver tissue pathological statistical results:After modeling, among the brain, heart and liver tissue pathology index, only liver tissue pathological index had significant difference (P< 0.01). After 1 week,2 weeks administration, compared with control group, pathological change scores in Danggui Buxue Decoction group, Xiaochaihu Decoction group, Chaihu Shengxue prescription group were significantly decreased(P<0.05).Conclusions:(1) The connotation of liver generating blood and Qi is that liver has the function of promoting blood generation due to its sprouted power, which will help the other viscera to produce blood. So we can regulate liver to promote blood generation in clinic treatment.(2) Chaihu Shengxue prescription can improve mice weight obviously. For hematogenesis effect, Xiaochaihu Decoction shows the early promoting effects in hematogenesis process and Xiaochaihu and Danggui Buxue compound Decoction shows the lasting hematogenesis effect. Chaihu Shengxue prescription has hematogenesis effect in the 2th week especially the WBC generation, which has a better effect than that of the other treatment decoction. Hematogenesis effect of Danggui Buxue Decoction is worse than that of xiaochaihu and Danggui Buxue compound Decoction and Chaihu Shengxue prescription.(3) Blood deficiency mice model prepared by complex etiology has little effect on heart and brain tissue pathology, In tissue blood deficiency, liver is sensitive than heart and brain. After administration, liver pathological ischemia were improved.(4)The mechanism of Xiaochaihu Decoction promoting hematogenesis process may be related to expression of SCF which is a early cytokine of HSC proliferation. Xiaochaihu Decoction, xiaochaihu and Danggui Buxue compound Decoction and Chaihu Shengxue prescription can significantly promote EPO expression, inhibit TGF-?1 and TNF-a expression, which can be explained by Chinese Medicine theory of liver generating blood and qi. Liver adjusts Qi and governs the generation Qi of spring.(5) The generation Qi of spring brings the power of birth and born, which can regulate the function of organs, boost the body upright, so as to effectively promote the functional activities of hematogenesis. Treatment of blood deficiency disease from liver is effective and feasible.This study provides a new way for the clinical treatment of anemia, and an experimental basis for liver generating blood and Qi.
