Therapeutic Effect And Mechanism Of Taohong Siwu Granule On Rats With Dysmenorrhea Due To Cold Coagulation And Blood Stasis

Objective:By establishing the model of dysmenorrhea of cold coagulation stasis type,the therapeutic effect of Taohong Siwu granule group on dysmenorrhea model rats was observed,and its molecular mechanism and the mechanism of its action on neuroendocrine system were discussed.Method:The first part:The dysmenorrhea rat model was established by ice water bath,oxytocin and estradiol valerate.The writhing reaction and the hemorheology were measured.The uterine morphology were observed.The contents of TXB2,6-keto-PF1?,ET,NO in serum and ET,NO,PGF_2?,PGE₂,AVP in uterine tissue were detected by ELISA.The concentration of Ca²⁺in uterine tissue were detected by confocal laser microscopy.The second part:Duplicate the rat model of dysmenorrhea.The contents of FSH,LH,E₂,P,?-EP in serum were detected by ELISA.ER,PR,OTR protein expression was detected by WB and immunohisto-chemistry,Gn RH,Gn RH-R protein and gene expression were detected by WB and RT-PCR.Results:1.Taohong Siwu granule can alleviate the writhing reaction of rats with cold coagulation,blood stasis and dysmenorrhea,reduce the number of writhing,the time of writhing and the incidence of writhing?P<0.05 or P<0.01?,The whole blood viscosity and plasma viscosity were decreased?P<0.05 or P<0.01?.It can thicken the uterine wall,obviously reduce the glands on the endometrium,and basically restore the normal state of the uterus;In addition,it also decreased the level of TXB₂,ET and increased the level of 6-keto-PGF_1?,NO in serum;decreased the level of PGF_2?,ET,AVP,Ca²⁺and increased the level of PGE₂?NO in uterine tissue?P<0.05 or P<0.01?;2.Taohongsiwu granule can reduce the content of FSH,LH and E₂,increase the content of P and?-EP?P<0.05 or P<0.01?in serum,decrease the protein expression of ER and OTR,increase the protein expression of PR?P<0.05 or P<0.01?,and decrease the protein and gene expression of Gn RH,Gn RH-R in hypothalamus and ovary?P<0.05 or P<0.01?.Conclusion:?1?Taohong Siwu granule can effectively relieve dysmenorrhea symptoms by regulating TXB₂?6-keto-PGF_1??PGF_2??PGE₂?ET?NO?AVP?Ca²⁺levels,inhibiting uterine contraction and increasing blood flow,which embodies the theory of"activating blood circulation and removing stasis"and"general principles without pain"in the treatment of dysmenorrhea in traditional Chinese medicine.?2?Taohong Siwu granule,through regulating HPOA related hormones and receptors,affects the neuroendocrine system,regulates the physiological and pathological processes related to dysmenorrhea,and embodies the systematic view of studying the pharmacology of traditional Chinese medicine.
